摘 要:目的探索卡介菌多糖核酸调控miR-155/SOCS1轴治疗特应性皮炎(AD)小鼠的作用机制。方法BALB/c小鼠随机分为对照组、模型组、卡介菌多糖核酸低剂量(0.05 mL/kg)组、卡介菌多糖核酸中剂量(0.1 mL/kg)组、卡介菌多糖核酸高剂量(0.15 mL/kg)组、地塞米松(0.5 mg/kg)组。除对照组外,其余组小鼠制备AD小鼠模型,分组处理后,观察各组小鼠耳部皮炎症状,并进行评分;苏木精-伊红染色(HE)检测各组小鼠耳组织病理情况;酶联免疫吸附法(ELISA)检测各组小鼠血清肿瘤坏死因子α(TNF-α)、白细胞介素-6(IL-6)、免疫球蛋白E(IgE)水平;实时荧光定量(qRT-PCR)检测各组小鼠耳组织miR-155、SOCS1表达;免疫印迹法检测各组小鼠耳组织SOCS1表达。结果与对照组比较,模型组小鼠耳组织出现特应性皮炎症状,皮炎评分、IgE及TNF-α、IL-6、miR-155水平显著升高(P<0.05),SOCS1表达降低(P<0.05)。与模型组比较,卡介菌多糖核酸低、中、高剂量组及地塞米松组小鼠耳组织皮炎症状减轻,皮炎评分、IgE及TNF-α、IL-6、miR-155水平降低(P<0.05),SOCS1表达升高且卡介菌多糖核酸各剂量之间有剂量依赖性(P<0.05)。卡介菌多糖核酸高剂量组与地塞米松组相比,差异无统计学意义(P>0.05)。结论卡介菌多糖核酸可下调miR-155表达、上调SOCS1表达,抑制炎症反应,改善AD小鼠的皮炎症状。Objective To explore the mechanism of BCG polysaccharide nucleic acid in the treatment of atopic dermatitis(AD)in mice by regulating miR-155/SOCS1 axis.Methods BALB/c mice were randomly divided into control group,model group,low dose of BCG polysaccharide nucleic acid(BCG-PSN)(0.05 mL/kg),medium dose of BCG polysaccharide nucleic acid(0.1 mL/kg),high dose of BCG polysaccharide nucleic acid(0.15 mL/kg)and dexamethasone(0.5 mg/kg)groups.Except for the control group,AD was established in mice in the other groups.The symptoms of ear dermatitis in each group were observed and scored.Hematoxylin-eosin staining(HE)was used to demonstrate the pathological changes in ear tissues.Enzyme-linked immunosorbent assay(ELISA)was employed to measure the serum levels of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and immunoglobulin E(IgE).Real-time fluorescence quantitative(qRT-PCR)was used to detect the levels of miR-155 and SOCS1 in the ear tissues.Western blotting was employed to detect the expression of SOCS1 protein in the ear tissues.Results AD in the ear tissue,the ear dermatitis score,IgE and TNF-α,IL-6,miR-155 level were significantly increased,while the expression of SOCS1 was significantly decreased in the model group compared with the control group.The symptoms of ear dermatitis were alleviated,the ear dermatitis score,IgE and TNF-α,IL-6,miR-155 levels were significantly decreased,and the expression of SOCS1 was significantly increased in BCG polysaccharide nucleic acid low,medium and high dose groups and dexamethasone group compared with the model group.Moreover,there was a dose-dependent relationship between the BCG polysaccharide nucleic acid groups(P<0.05).There was no significant difference between BCG-PSN high dose group and the dexamethasone group(P>0.05).Conclusion BCG polysaccharide nucleic acid can down-regulate the expression of miR-155,up-regulate the expression of SOCS1,inhibit inflammation,and improve the dermatitis symptoms in AD mice.
