摘 要:目的:探讨杜仲总黄酮对多囊卵巢综合征(polycystic ovarian syndrome,PCOS)模型大鼠的干预作用。方法:空白组除外,采用颈背部皮下注射脱氢表雄酮(dehydroepiandrosterone,DHEA)溶液0.2 mL(0.06 mg·g^-1)法建立PCOS大鼠模型。模型大鼠随机分为模型组,达英35组(0.3392 mg·kg^-1),杜仲总黄酮高剂量组、中剂量组、低剂量组(200、100、50 mg·kg^-1)。各组动物每天给药1次,连续给药21 d后取材,计算体质量、卵巢指数、子宫指数。酶联免疫吸附法测定各组大鼠血清中雌二醇(estradiol,E2)、睾酮(testosterone,T)、促性腺激素释放激素(gonadotrophin releasing hormone,GnRH),卵泡刺激素(follicle-stimulating hormone,FSH)、催乳素(prolactin,PRL)、促黄体生成素(luteinizing hormone,LH)、17α-羟孕酮(17α-hydroxyprogesterone,17α-OHP)、孕酮(progesterone,P)、胰岛素(insulin,INS)水平。结果:与模型组比较,杜仲总黄酮各剂量组可不同程度降低模型大鼠体质量、子宫及卵巢相关指数,降低模型动物血清中GnRH、LH、T、PRL、INS、17α-OHP水平,升高模型动物血清中FSH、E2、P水平(P<0.05或P<0.01)。杜仲总黄酮低剂量组可降低模型动物血清中INS水平,但与模型组比较,差异无统计学意义(P>0.05)。结论:杜仲总黄酮可有效降低多囊卵巢综合征大鼠体质量、卵巢指数、子宫指数,降低模型动物血清中GnRH、LH、T、PRL、INS、17α-OHP水平,升高模型动物血清中FSH、E2、P水平。Objective:To investigate the intervention effect of eucommia ulmoides flavonoids on polycystic ovarian syndrome(PCOS)model rats.Methods:Except the blank group,the PCOS rat model was established by subcutaneous injection of 0.2 mL(0.06 mg·g^-1)of Dehydroepiandrosterone(DHEA)solution in the back of the neck.Model rats were randomly divided into model groups,Daying 35 group(0.3392 mg·kg^-1),eucommia ulmoides high-dose group,middle-dose group,and low-dose group(200,100,50 mg·kg^-1).Animals in each group were administered once a day,and samples were taken after 21 days of continuous administration to calculate body mass,ovarian index,and uterine index.Enzyme-linked immunosorbent assay for the determination of estradiol(E2),testosterone(T),gonadotrophin releasing hormone(GnRH),follicle-stimulating hormone(follicle-stimulating hormone,FSH),prolactin(PRL),luteinizing hormone(LH),17α-hydroxyprogesterone(17α-OHP),progesterone(P),insulin(INS)Level.Results:Compared with the model group,each dose group of total flavonoids of eucommia ulmoides can reduce the body weight,uterus and ovary-related indexes of model rats to different degrees,reduce the levels of GnRH,LH,T,PRL,INS,17α-OHP in the serum of model animals while raise FSH,E2,P levels in serum of high model animals(P<0.05 or P<0.01).The low-dose group of total flavonoids in eucommia ulmoides can reduce the INS level in the serum of model animals,but compared with the model group,the difference was not statistically significant(P>0.05).Conclusion:The total flavonoids of eucommia ulmoides can effectively reduce the body mass,ovarian index and uterine index of polycystic ovary syndrome rats,reduce the levels of GnRH,LH,T,PRL,INS,17α-OHP in the serum of model animals and increase the serum of model animals FSH,E2,P level.
