摘  要:目的:探讨曲古抑菌素A对膀胱癌细胞凋亡及细胞周期进程的影响及机制。方法:使用5637细胞(购自上海中国科学院细胞所)作为研究细胞,使用含有低、中、高浓度曲古抑菌素A的RPMI 1640培养基培养5637细胞24、48、72 h。然后检测各组细胞相对存活率、凋亡率、处于各细胞周期阶段的细胞比例以及细胞周期相关蛋白表达。采用方差检验和 t检验。 结果:随着药物浓度升高和药物作用时间延长,5637细胞相对生存率[空白组:100.00%、100.00%、100.00%;低剂量组:(90.45±18.56)%、(80.45±12.25)%、(65.45±8.65)%;中剂量组:(71.02±9.56)%、(63.45±6.12)%、(50.42±6.35)%;高剂量组:(52.14±6.25)%、(42.11±5.14)%、(20.25±3.25)%]( F=11.483、9.244、8.264, P<0.01),细胞凋亡率显著升高;随着药物浓度增加处于亚G 1期细胞所占比例显著升高,p21蛋白显著升高,磷酸化蛋白激酶B(p-Akt)及细胞周期蛋白D显著降低。低剂量组和中剂量组细胞处于S期细胞比例显著低于空白组。 结论:曲古抑菌素A可以通过抑制p-Akt信号通路促进膀胱癌细胞凋亡同时也可以通过促进p21蛋白表达抑制细胞周期进程。Objective To explore the effect and mechanism of trichostatin 1 on apoptosis and cell cycle of bladder cancer cells.Methods 5637 cells were used as study cells,and RPMI 1640 medium containing low,medium and high concentrations of trichostatin 1 was used to culture 5637 cells for 24,48 and 72 hours.Then the relative survival rate,apoptosis rate,the proportion of cells in each cell cycle stage and the expression of cell cycle related proteins were detected.Results With the increase of drug concentration and the prolongation of drug action time,the relative survival rate of 5637 cells[blank group:100.00%,100.00%,100.00%;low dose group:(90.45±18.56)%,(80.45±12.25)%,(65.45±8.65)%;medium dose group:(71.02±9.56)%,(63.45±6.12)%,(50.42±6.35)%;high dose group:(52.14±6.25)%,(42.11±5.14)%,(20.25±3.25)%,F=11.483,9.244,8.264,P<0.01],the apoptosis rate of cells in the sub G1 phase increased significantly phosphorylated-protein kinase B(p-Akt)and Cyclin D decreased significantly.The proportion of cells in S phase in low dose group and middle dose group was significantly lower than that in control group.Conclusion Trichostatin 1 can promote apoptosis of bladder cancer cells by inhibiting p-Akt signal pathway,and also can inhibit cell cycle by promoting p21 protein expression.
