摘 要:目的研究不同氧浓度下人造血干细胞(HSCs)生长及凋亡状况的差异,探究不同氧浓度对HSCs生长的抑制作用。方法将HSCs分别在不同氧浓度下培养,3%、5%、15%氧浓度作为实验组,20%氧浓度作为对照组。分别培养4、7、10 d,研究红系集落形成单位(CFU-E)集落数;分别培养6、12、24 h,观察HSCs细胞凋亡情况。结果培养4、7、10 d,5%氧浓度组CFU-E集落数最高,3%和5%氧浓度组与对照组CFU-E集落数比较,差异均有统计学意义(P<0.05)。培养6 h,5%氧浓度组HSCs凋亡率最低,3%和5%氧浓度组与对照组HSCs凋亡率比较,差异均有统计学意义(P<0.05);培养12 h,5%氧浓度组HSCs凋亡率最低,5%和15%氧浓度组与对照组HSCs凋亡率比较,差异均有统计学意义(P<0.05);培养24 h,5%氧浓度组HSCs凋亡率最低,3%和5%氧浓度组与对照组HSCs凋亡率比较,差异均有统计学意义(P<0.05)。培养4、7、10 d,氧浓度变动与CFU-E集落数均呈正相关(P<0.05)。培养6、12、24 h,氧浓度变动与HSCs凋亡率均呈正相关(P<0.01)。结论不同氧浓度对HSCs的生长和凋亡有直接影响。5%的氧浓度能促进HSCs生长,同时又能抑制HSCs凋亡。Objective To study the differences in the growth and apoptosis of hematopoietic stem cells(HSCs)under different oxygen concentrations,and to explore the inhibitory effect of various oxygen concentrations on the growth of HSCs.Methods HSCs were cultured at a variety of different oxygen concentrations,3%,5%and 15%oxygen concentration were taken as the experimental group,and 20%oxygen concentration was taken as the control group.The number of erythroid colony forming unit(CFU-E)clusters was studied after 4,7 and 10 days.HSCs were cultured for 6,12 and 24 hours respectively for observing the apoptosis of HSCs cells.Results After 4,7,10 days of cultivation,5%oxygen concentration group had the highest number of CFU-E colonies,and the differences between 3%and 5%oxygen concentration group and control group were statistically significant(P<0.05).After cultivation for 6 hours,the HSCs apoptosis rate in 5%oxygen concentration group was the lowest,and the differences between 3%and 5%oxygen concentration group and control group were statistically significant(P<0.05).5%oxygen concentration group had the highest number of cfu-e colonies,and the difference between 3%group and control group was statistically significant(P<0.05).After cultivation for 12 hours,the apoptosis rate of HSCs in 5%oxygen concentration group was the lowest,and the differences between 5%and 15%oxygen concentration group and control group were statistically significant(P<0.05).After cultivation for 24 hours,the HSCs apoptosis rate in 5%oxygen concentration group was the lowest,and the differences between 3%and 5%oxygen concentration group and control group were statistically significant(P<0.05).4,7 and 10 days after cultivation,the changes of oxygen concentration were positively correlated with the results of CFU-E colony number(P<0.05).After 6,12 and 24 hours of cultivation,the changes of oxygen concentration was positively correlated with the apoptosis rate of HSCs(P<0.01).Conclusion The growth and the apoptosis of HSCs are directly influenced by differe
