进境日本圆柏上亚洲胶孢锈病菌的检疫鉴定

摘　　要：自宁波口岸进境日本圆柏枝叶上,采集获得3份胶锈菌属的标本,编号为JC1、JC2、JC3,采用形态学观察和分子鉴定方法对上述样品进行了研究。形态学观察确认3份标本上的锈菌属于胶锈菌,仅有冬孢子阶段的形态特征,冬孢子堆丛枝状,生长在枝干上,但不产生明显的隆起,2~6 mm高,棕褐色;冬孢子双胞,圆柱形至拟纺锤形或椭圆形,31~50μm×16~27μm,壁厚1.0~2.5μm,桔黄色,芽孔1~2个,位于近膈膜处;孢柄无色,圆柱形,直径2.5~5.0μm。通过对rDNA 18S,ITS,28S序列测定,3份标本上锈菌序列互相之间具有100%的相似度。构建系统发育分析进一步发现3份标本与GenBank中多株亚洲胶孢锈病菌聚在同一分支上。结合形态及分子系统发育分析结果,明确采集于日本圆柏上所截获病菌为亚洲胶孢锈病菌,这是我国口岸首次截获该种重要植物病原菌。Three rust fungal specimens(JC1,JC2 and JC3) were obtained from Juniperus chinensis imported from Japan. Although only telial stage was found,identification based on morphological and molecular phylogenetic analyses assigned them to the genus Gymnosporangium. Their telia were found on green stems,developing on witche’s broom without causing swelling on stem,2-6 mm high,brownish;teliospores2-celled,cylindrical to spindle or ellipsoid,31-50 ×16-27 μm,walls 1.0-2.5 μm,orange,pores 1 or 2 near septum;pedicels cylindrical,hyaline,2.5-5.0 μm in diameter. Sequence data of rDNA 18S,ITS and 28S of three specimens had 100% similarities and molecular phylogenetic studies revealed that they were clustered with several specimens of Gymnosporangium asiaticum deposited in GenBank. Both morphological and molecular evidences confirmed the identity of this rust species on Junipe rus chinensis as G. asiaticum. To our knowledge,this is the first report for interception of G. asiaticum in China customs.
