贵州民间草药血盆草止血活性药效初步研究

摘　　要：目的:探讨贵州民间草药血盆草的止血药效及活性部位。方法:采用中药造模的方法建立小鼠出血模型,醇提物灌胃给药7天后用剪尾法和毛细玻管法测定出血、凝血时间,并统计小鼠血小板数,评价血盆草醇提物的止血作用;采用经典的系统溶剂法(极性由小到大:石油醚-氯仿-乙酸乙酯-正丁醇)萃取血盆草的95%乙醇提取物,经中药造模方法建立小鼠出血模型,灌胃给药7天后,取眼眶血离心后取血浆检测小鼠血浆的TT、PT、APTT及Fib浓度,综合分析全面评价,确定血盆草的止血药效活性部位。结果:模型用药组的出血时间、凝血时间均缩短,血小板数升高;凝血四项检测中,与空白组比较,模型组小鼠的TT、PT、APTT均缩短,Fib浓度降低,具极显著性差异(P<0.01);与模型组比较,各组出血性小鼠的TT、PT、APTT均缩短,Fib浓度升高,石油醚和氯仿组具极显著性差异(P<0.01)。结论:血盆草有较好的止血药效,其活性部位为石油醚和氯仿部位;检测方法简便、可靠。Objective:To investigate the hemostatic effect and active site of Salvia cavaleriei var.simplicifolia Stib.Methods:The bleeding model of mice was established by the method of traditional Chinese medicine.The bleeding and clotting time of mice were measured by tail clipping and capillary glass tube methods after 7 days of gastric administration of alcohol extract,and the platelet count of mice was counted to evaluate the hemostatic effect of alcohol extarct of the plant.The classical system solvent method(small to large:petroleum ether-chloroform-ethyl acetate-n-butanol)was used to extract the alcohol extract of the plant,and the mouse bleeding model was established by the traditional Chinese medicine modeling method.After 7 days of drug administration,the concentration of TT,PT,APTT and Fib in the plasma of the mice was determined by centrifugation of the orbital blood,and comprehensive analysis was performed to determine the hemostatic activity site of the plant.Results:The bleeding time and clotting time were shortened and the number of platelets was increased.In the four coagulation tests,TT,PT and APTT of the model group were shortened and Fib concentration was reduced compared with that of the blank group,with extremely significant difference(P<0.01).Compared with model group,TT,PT and APTT of hemorrhagic mice in each group were shortened,Fib concentration was increased,and petroleum ether and chloroform groups showed extremely significant differences(P<0.01).Conclusion:The blood pellucida has good hemostatic effect,and its active site was petroleum ether and chloroform extracts.The detection method is simple and reliable.
