摘 要:目的探讨NUMB对胆囊癌细胞NOZ和GBC-SD的增殖和迁移的作用。方法应用RNA干扰技术敲低人胆囊癌细胞NOZ和GBC-SD中NUMB的表达,将人胆囊癌细胞NOZ和GBC-SD分别分为实验组(si-NUMB组)和对照组(CTRL组),其中si-NUMB组转染si-NUMB,CTRL组转染阴性对照剂si-NC。采用CCK-8法检测两组细胞的增殖能力[吸光度(OD)值来表示];采用Transwell小室实验检测两组细胞的迁移能力;采用Western blot法检测NUMB及上皮间质转化关键蛋白的表达水平。结果NOZ细胞中,与CTRL组相比,si-NUMB组中的OD值在第2、3、4、5天时均明显为高(均P<0.05);GBC-SD细胞中,与CTRL组相比,si-NUMB组中的OD值在第3、4、5天时均明显为高(均P<0.05)。与CTRL组比较,si-NUMB组中人胆囊癌NOZ和GBC-SD细胞的迁移细胞的数量均明显升高(均P<0.05)。与CTRL组相比,NOZ和GBC-SD细胞在si-NUMB组中NUMB、E-cadherin蛋白表达水平均明显为低而Snail-1、N-cadherin蛋白表达水平均明显为高(均P<0.01)。结论NUMB能够抑制胆囊癌细胞的增殖和迁移能力,可能作为胆囊癌治疗的靶标。Objective To investigate the effect of NUMB gene on proliferation and migration of gallbladder carcinoma cells.Methods RNA interference technology was used to knock down NUMB expression in gallbladder carcinoma NOZ and GBC-SD cells.The si-NUMB and si-NC were transfected into NOZ and GBC-SD cells of study group and control group,respectively.CCK-8 assay was performed to evaluate the proliferation of NOZ and GBC-SD cells;Transwell migration assay was used to detect the migration of NOZ and GBC-SD cells;the changes of EMT-related proteins were detected by Western blot assay.Results The proliferation effect of si-NUMB group was significantly higher than that of CTRL group at 2,3,4 and 5 days in NOZ cells as well as which at 3,4 and 5 days in GBC-SD cells(P<0.05).The Migration effect of NOZ cells and GBC-SD cells of human gallbladder cancer in the si-NUMB group was significantly higher than that in the CTRL group(P<0.05).After transfection with si-NUMB,the expressions of Snail-1 and N-cadherin were significantly increased,while the expression of E-cadherin was significantly decreased(all P<0.05).Conclusion NUMB can promote the proliferation and migration of gallbladder carcinoma cells,indicating that it may be used as a therapeutic target for gallbladder cancer.
