摘 要:目的观察去甲斑螯素对去势骨质疏松症小鼠骨微结构的影响,探讨其防治骨质疏松症的可行性。方法40只雌性C57BL-6J 2月龄小鼠随机分为低剂量组、高剂量组、假手术组和去卵巢组,每组10只。除假手术组外,其它3组均通过摘除双侧卵巢建立小鼠骨质疏松模型。建模8周后,各组每日对应给予腹腔注射相应浓度的去甲斑螯素溶液或0.9%氯化钠注射液,连续注射4周。处死各组小鼠后用显微CT检查分析小鼠股骨远端骨小梁骨密度、骨体积分数(BV/TV)、骨小梁数量(Tb.N)、骨小梁厚度(Tb.Th)和骨小梁间隔(Tb.Sp)。结果去卵巢组骨小梁骨密度、BV/TV、Tb.N、Tb.Th均低于假手术组,Tb.Sp高于假手术组(均P<0.05);低剂量组与高剂量组骨小梁骨密度、BV/TV、Tb.N、Tb.Th均高于去卵巢组,Tb.Sp均低于去卵巢组(均P<0.05)。结论去甲斑螯素能有效减少卵巢摘除引起的骨量丢失,具有剂量依赖性,是一种治疗骨质疏松症的一种潜在选择。Objective To explore the effects of norcantharidin on bone microstructure of ovariectomy-induced osteoporosis rats.Methods Forty female C57 BL-6 J mice of two-month-old were divided into four groups:sham-operated group(Sham),model group,low dose norcantharidin group and high dose norcantharidin group with 10 mice in each group.The osteoporosis model was established by resection of bilateral ovaries in model group and two treatment groups.Eight weeks after modeling,the norcantharidin or normal saline were given by intraperitoneal injection for 4 weeks in corresponding groups.Then the mice were sacrificed,and the bone mineral density,bone volume fraction(BV/TV),trabecular number(Tb.N),trabecular thickness(Tb.Th)and trabecular separation(Tb.Sp)of the distal femur were analyzed by micro-CT.Results Micro-CT showed that bone mineral density,BV/TV,Tb.N,Tb.Th were significantly higher than that in sham in ovariectomized mice(all P<0.05).While the Tb.Sp was significantly lower than those in sham group.In low dose group and high dose group,bone mineral density,BV/TV,Tb.N and Tb.Th increased and Tb.Sp decreased compared to model group(al P<0.05).Conclusion Norcantharidin can effectively inhibit bone loss and is an ideal treatment for osteoporosis.
