肠乐一号灌肠液对溃疡性结肠炎模型大鼠白细胞介素-1β的影响

摘　　要：目的:观察肠乐一号灌肠液对溃疡性结肠炎(Ulcerative Colitis,UC)模型大鼠白细胞介素-1β(interleukin-1β,IL-1β)的影响。方法:将60只SD大鼠按照随机数字表法分为正常组、模型对照组、中药组和阳性对照组,每组各15只。除正常组外,其余各组大鼠采用三硝基苯磺酸法造模。造模后中药组给予肠乐一号灌肠液灌肠给药,每日1次;阳性对照组给予美沙拉嗪溶液灌肠,每次2 mL,每日1次;模型对照组给予等体积的生理盐水灌肠。观察大鼠造模前、给药后一般情况、体质量及结肠黏膜组织病理变化,检测各组大鼠血清及结肠组织中IL-1β含量。结果:中药组结肠黏膜损伤程度评分低于模型对照组与正常组,差异具有统计学意义(P<0.05),中药组结肠黏膜损伤程度评分低于阳性对照组,但差异无统计学意义(P>0.05)。中药组血清及结肠组织中IL-1β含量均低于模型对照组与正常组,差异具有统计学意义(P<0.05)。血清及结肠组织中IL-1β含量与阳性对照组比较,差异无统计学意义(P>0.05)。结论:肠乐一号灌肠液能改善溃疡性结肠炎模型大鼠结肠黏膜组织的损伤程度,降低血清中IL-1β的含量,下调结肠组织中IL-1β表达,从而修复溃疡。Objective:To observe the influence of NO.1 Enema Decoction for Intestines on interleukin-1β(IL-1β)in rats with ulcerative colitis(UC).Methods:A total of 60 SD rats were randomly divided into the normal group,the model control group,the TCM group and the positive control group,with 15 rats in each group.Except the normal group,the other rats were modeled by trinitrobenzene sulfonic acid method.After modeling,the TCM group was given NO.1 Enema Decoction for Intestines,once a day;the positive control group was given mesalazine solution for enema,2 mL each time,once a day;the model control group was given equal volume of normal saline enema.The changes of general condition,body weight and pathological colonic mucosa were observed before modeling and after administration.The contents of IL-1βin serum and colon tissue were detected.Results:The score of colonic mucosal injury in the TCM group was lower than that in the model control group and normal group,and the difference was statistically significant(P<0.05).The score of colonic mucosal injury in the TCM group was lower than that in the positive control group,but the difference was not statistically significant(P>0.05).The contents of IL-1βin serum and colon tissue of the TCM group were all lower than those of the model control group and normal group,and all the differences were statistically significant(P<0.05).There was no significant difference in the contents of IL-1βin serum and colon tissue between the TCM group and the positive control group(P>0.05).Conclusion:NO.1 Enema Decoction for Intestines can improve the damage degree of colonic mucosa in rats with UC,reduce the content of IL-1βin serum,and down-regulate the expression of IL-1βin colon tissue,so as to repair the ulcer.
