摘 要:测定了桑树桑黄(Sanghuangporus sanghuang)10个菌株发酵菌丝体的生物量、多糖、黄酮、三萜及多酚含量,并比较其对DPPH自由基、羟自由基的清除能力和总还原力,分析活性成分含量与抗氧化能力的相关性。结果表明:菌株SH06发酵菌丝体的黄酮和多酚含量较高,抗氧化能力较强,且黄酮含量与3个抗氧化指标显著相关,表明黄酮是桑树桑黄抗氧化作用的主要活性物质。比较了萘乙酸(NAA)、细胞分裂素(6-BA)、3-吲哚乙酸(IAA)、水杨酸(SA)、脱落酸(ABA)这5种植物激素对菌株SH06液体发酵的影响,发现发酵培养基中添加10 mg·L^-1 6-BA可将菌丝体生物量及胞内黄酮含量分别提高1.38和1.76倍。Ten strains of Sanghuangporus sanghuang were cultivated by liquid fermentation,then measured for total polysaccharides,total flavonoids,total triterpenes and total polyphenols in their mycelia,and also determined for DPPH free radical scavenging ability,hydroxyl radical scavenging capacity and total reducing power to establish a correlation between active ingredient content and antioxidant capacity.The results showed that strain SH06 had higher flavonoid and polyphenol content and stronger antioxidant capacity than the rest 9 strains.Correlation analysis showed that flavonoid content was significantly correlated with antioxidant capacity,indicating that flavonoids are the main group of active substances contributing to the antioxidant activity of S.sanghuang.In order to obtain greater flavonoid content,SH06 was elicited during liquid fermentation by five plant hormones,including naphthaleneacetic acid(NAA),6-benzylaminopurine(6-BA),3-indoleacetic acid(IAA),salicylic acid(SA),and abscisic acid(ABA).It was found that 10 mg·L^-1 6-BA increased the biomass and flavonoid content of mycelium by 1.38 and 1.76 times,respectively.
