摘 要:目的对人群尿液测定技术(石墨炉原子吸收法)进行推广,研制一般人群尿钴正常值,为硬金属肺病诊断生物标志物的制定提供线索。方法使用磷酸氢二铵基体改进剂,采用石墨炉原子吸收法直接进样,测定尿钴浓度,提高灰化温度,减少原子化阶段的气相干扰,并探讨最佳分析条件。结果磷酸氢二铵作为基体改进剂吸光度最大为0.555,最佳灰化温度1200℃和原子化温度2200℃,检出限为0.448μg/L,回收率范围为96.54%~107.38%,批内精密度范围为0.93%~2.30%。124名研究对象中尿钴最低检出值为0.470μg/L,最大值为6.820μg/L,检出限为0.448μg/L,其中有16名未检出,检出率为87.09%。全距为0.448~6.820μg/L,M(P25,P75)为2.790(1.697,4.300)μg/L,均数为3.037μg/L,单侧估计95%上限为6.170μg/L,99%上限为6.790μg/L。一般健康成年人尿钴含量的范围在0.000~6.170μg/L。结论本实验摸索建立以石墨炉原子吸收光谱法测定尿中钴含量的方法可行,具有尿样无需繁杂预处理,简便快速、准确可靠,适用于人群大批量尿样的检测等优点,可应用于临床检测,结果准确,是检测尿中钴含量的简单可行方案。可为今后硬金属肺病诊断中生物标志物(尿钴生物接触限值)的制定提供实验室测定方法和数值参考。Objective To popularize the technique of urine determination[graphite furnace atomic absorption spectrometry(GFAAS)],develop the normal value of urine cobalt in general population,and provide clues for the development of biomarkers for the diagnosis of hard metal lung disease.Methods Using diammonium hydrogen phosphate as matrix modifier and direct injection with the graphite furnace atomic absorption method,the concentration of cobalt in urine was measured.The ashing temperature was increased and gas phase interference in atomization stage was reduced,and the optimum analysis conditions were discussed.Results The maximum absorbance of diammonium hydrogen phosphate as matrix modifier was 0.555.The optimum ashing temperature was 1200℃and atomization temperature was 2200℃.The detection limit was 0.448μg/L,the recovery range was 96.54%-107.38%,and the intra batch precision range was 0.93%-2.30%.The lowest detection value of urinary cobalt in 124 subjects was 0.470μg/L,the maximum value was 6.820μg/L,and the detection limit was 0.448μg/L,of which 16 were not detected,the detection rate was 87.09%.The total distance was 0.448-6.820μg/L,M(P25-P75)was 2.790(1.697,4.300)μg/L,and the mean was 3.037μg/L.The unilateral upper limit of 95%was 6.170μg/L,and the upper limit of99%was 6.790μg/L.The range of urinary cobalt content in healthy adults was 0.000-6.170μg/L.Conclusion The method of determination of cobalt in urine by graphite furnace atomic absorption spectrometry is feasible,has the advantages of no need of complicated pretreatment for urine sample,simple,rapid,accurate and reliable,and suitable for the detection of large quantities of urine samples.The method can be applied to clinical detection with accurate results,and it is a simple and feasible scheme for the detection of cobalt in urine.It can provide laboratory test method and numerical reference for the establishment of biomarkers(urinary cobalt biological exposure limit)in the diagnosis of hard metal lung disease in the future.
