摘　　要：目的探究IL-6对呼吸系统趋化因子表达谱及免疫细胞组成的影响。方法通过real-time PCR对IL-6敲除小鼠的上呼吸道鼻黏膜和下呼吸道肺组织中56种趋化因子及受体的表达谱进行芯片分析,利用流式细胞术对上下呼吸道中免疫细胞组成进行分析;再使用IL-6蛋白对野生型小鼠进行滴鼻处理,检测鼻黏膜和肺组织内趋化因子表达谱和免疫细胞组成的改变。结果IL-6敲除后小鼠肺组织CXCL5表达显著降低,鼻黏膜中T细胞占比升高,肺组织中肺泡巨噬细胞和CD4^+T细胞占比均降低;IL-6滴鼻后鼻黏膜中CCL3、CXCL2和CXCL3的表达显著升高,肺组织中总白细胞占比显著减少,除中性粒细胞占比增加外,其它免疫细胞均有减少。结论降低或升高IL-6水平均可导致呼吸系统内多种趋化因子表达以及免疫细胞组成发生改变,提示IL-6可调节呼吸系统免疫,尤其是中性粒细胞的免疫反应。This study aims to investigate the effect of interleukin(IL)-6 on respiratory chemokine levels and immune cells.In the nasal and lung of IL-6 deficient mice,56 chemokines and chemokine receptors were detected by real-time PCR to investigate the expression variation induced by IL-6 deficiency,while flow cytometry of immune cells was performed to study the effect of IL-6 on the movement of immune cells.Otherwise,the same analysis was employed in IL-6 instilled mice.Data showed that deficiency of IL-6 resulted in the downregulation of CXCL5,alveolar macrophage and CD4^+T cells in lung,while T cells increase in nasal.After IL-6 instillation,the expression of CCL3,CXCL2 and CXCL3 increased in nasal,the proportion of total leukocytes in lung decreased,while neutrophils increased.In conclusion,the change of IL-6 level could alter the expression of chemokines and movement of immune cells in respiratory system,which indicates that IL-6 can regulate immune responses in mouse respiratory system,especially the immune response of neutrophils.
