摘 要:目的探讨雷公藤内酯醇对支气管哮喘小鼠血浆IgE、BALF中炎症因子、BALF液中细胞总数及细胞分类变化以及肺组织病理变化的影响,评估该药物对支气管哮喘小鼠气道炎症反应及气道重塑干预过程可能的机制及作用,明确雷公藤内酯醇对支气管哮喘小鼠的临床应用价值。方法40只Balb/c雌性小鼠分为模型组、正常组、低剂量TP治疗组、高剂量TP治疗组,用OVA致敏的方法建立哮喘模型;低剂量组、高剂量按100μg/kg、200μg/kg腹腔注射TP溶液。检测各组小鼠肺组织病理改变、血浆IgE、BALF中炎症细胞、炎症因子表达情况。结果运用TP治疗的小鼠其支气管周围和血管周围炎症浸润程度减轻,TP可抑制血浆IgE水平,减少BALF中的炎症细胞数及其比例和炎症因子的上升。结论推测TP可能通过降低血浆IgE水平,减轻相应炎症因子及减少气道炎性细胞的浸润,尤其是对Eos的抑制效应,从而达到治疗哮喘的目的。To explore the possible mechanism and the clinical application value of triptolide(TP)in bronchial asthmatic mice treatment,we evaluated the effects of TP on the level of serum IgE,inflammation factors,the total number and cell classification in BALF,and the pathological changes of lung tissue in asthmatic mice.Total of 40 Balb/c mice were recruited and divided into model group,normal group,low dose TP-treatment group,and high dose TP-treatment group.The asthma model was established by using the method of OVA sensitization.Low dose TPtreatment group and high dose TP-treatment group were respectively treated with intraperitoneal injection of 100μg/kg and 200μg/kg.We detected the changes of lung tissue pathology,the levels of serum IgE,the numbers and ratios of inflammatory cells as well as the inflammatory factors in each group.TP-treatment groups can reduce the degrees of inflammation around the bronchi and blood vessels,and also reduce the levels of serum IgE,the numbers of inflammatory cells and inflammatory factors.In conclusion,TP may achieve the purpose of treating asthma by lowering the levels of serum IgE,reducing correspondingly inflammatory factors and inflammatory cells infiltration,especially inhibiting Eos.
