养任调冲汤改善更年期综合征大鼠糖脂代谢的机制研究

摘　　要：目的:观察养任调冲汤对更年期阴虚内热证大鼠糖脂代谢的影响并探讨其机制。方法:以去卵巢结合灌服热性中药的方法制备更年期阴虚内热证大鼠模型,养任调冲汤灌胃给药后,观察大鼠24 h饮水量、肛温、体质量增长等一般情况,检测血清甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)、空腹血糖(FPG)及空腹胰岛素(FINS)水平,计算稳态胰岛素抵抗指数(HOMAIR)并测定血清瘦素(LP)及脂联素(APN)水平。结果:养任调冲汤治疗后,大鼠体质量增长缓慢、饮水量减少、肛温下降;血清HDL显著升高,LDL、TC与TG水平显著降低;FINS、FPG、HOMAIR水平则差异无统计学意义。高、中剂量组血清APN水平有所增高。血清LP水平虽有所下降,但差异无统计学意义。结论:养任调冲汤能缓解更年期综合征大鼠的阴虚内热证,调节其血脂代谢,对改善更年期综合征可能有效,其作用机制可能与升高血清APN和降低LP水平有关。Objective:To observe the effect of Yangren Tiaochong Decoction on glucose and lipid metabolism and expound the mechanism in climacteric syndrome rats combined with yin deficiency and internal heat.Methods:The specific pathogen free(SPF)SD female rats were removed the bilateral ovaries combinated with perfusion of hot Chinese medicine for establishing the models of climacteric syndrome combined with yin deficiency and internal heat and was intragastrically given Yangren Tiaochong Decoction.The water intake,rectal temperature and weight were observed.Besides,the fasting blood glucose(FPG),fasting insulin(FINS),total cholesterol(TC),triglycerides(TG),high density lipoprotein(HDL)and low density lipoprotein(LDL)were determined,and homeostasis model assessment of insulin resistance index(HOMAIR)was calculated.Not only that,the levels of serum leptin(LP)and adiponectin(APN)were also measured.Results:After Yangren Tiaochong Decoction treatment,the water intake reduced,the rectal temperature declined,and the weight gain slackened.Moreover,the levels of TC,TG,LDL decreased,and the levels of HDL significantly increased in the climacteric syndrome rats combined with yin deficiency,while there were no significant changes in the level of FPG,FINS and HOMAIR.In addition,the serum APN level of Yangren Tiaochong Decoction treatment group was higher than the control group,and LP level was lower than the control group,but there was no statistical significance from differences between groups.Conclusion:Yangren Tiaochong Decoction can alleviate Yin deficiency and internal heat syndrome and has a better intervention on lipid metabolism disorders of rats with climacteric syndrome,and the mechanism may be associated with increasing serum APN and decreasing LP level.
