摘　　要：目的:探讨Duffy血型基因多态性和结直肠癌的关系。方法:收集325例结直肠癌患者和306例健康对照人群血样,应用TaqMan-MGB探针实时PCR方法进行Duffy血型的基因分型。结果:在对照组人群中,基因型FY*A/FY*A为266例(86.9%),FY*A/FY*B为39例(12.7%),FY*B/FY*B为1例(0.3%)。在结直肠癌患者中,基因型FY*A/FY*A为286例(88.0%),FY*A/FY*B为38例(11.7%),FY*B/FY*B为1例(0.3%)。结直肠癌组和对照组比较,Duffy血型基因型频率分布差异无统计学意义(P>0.05)。在淋巴结转移阴性的结直肠癌患者组FY*A/FY*B和FY*B/FY*B组合的基因型频率(16.7%)高于淋巴结转移阳性组的基因型频率(8.6%),而FY*A/FY*A的基因型频率(83.3%)要低于淋巴结转移阳性组的基因型频率(91.4%)(P<0.05)。基因型FY*A/FY*A的结直肠癌患者要比非FY*A/FY*A基因型的患者发生淋巴结转移的风险高2.137倍。结论:Duffy血型基因多态性与结直肠癌发生无关,与结直肠癌的淋巴结转移有关。Objective:To investigate the association of Duffy blood group gene polymorphism with colorectal cancer(CRC). Method:Blood samples from patients with CRC and control group were collected and genotyped using real-time PCR with TaqMan-MGB probes. Result:The numbers of FY*A/FY*A,FY*A/FY*B and FY*B/FY*B genotype were 266(86.9%),39(12.7%)and 1(0.3%)respectively in control group and 286(88.0%),38(11.7%)and 1(0.3%)respectively in CRC.There were no significant differences(P>0.05)in the genotype frequency distribution for Duffy blood gene between the patients and the control groups.Combination of FY*A/FY*B and FY*B/FY*B genotype frequencies(16.7%)in lymph node negative patients was higher than that in lymph node positive patients(8.6%).However,FY*A/FY*A genotype frequency(83.3%)in lymph node negative patients was lower than that in lymph node positive patients(91.4%)(P<0.05).CRC patients with FY*A/FY*A genotype had higher lymph node metastasis risk(2.137-fold)than patients without FY*A/FY*A genotype.Conclusion:Duffy blood group gene polymorphism is not associated with CRC tumorigenesis but is correlated with lymph node metastasis.
