摘  要:目的研究L-谷氨酰胺对急性酒精肝损伤的作用以及对核因子-κB(NF-κB)p65(NF-κB p65)表达的影响。方法按照体重将雄性小鼠随机分为3组:正常组、模型组和实验组,每组15只。实验组用L-谷氨酰胺800 mg·kg^-1·d^-1连续灌胃7 d,正常组和模型组均灌胃等体积盐水。给药7 d后,一次性灌胃乙醇12 mL·kg^-1后,诱导小鼠急性肝损伤。以全自动生化分析仪检测谷草转氨酶(GOT)和谷丙转氨酶(GPT)活性,计算小鼠的肝指数,用蛋白质印迹法测定各组小鼠肝组织中NF-κB p65蛋白的相对表达量(相对灰度值)。结果正常组、模型组和实验组的GOT分别为(113.9±11.4),(203.6±22.1)和(158.6±16.8)U·L^-1;这3组的GPT分别为(52.8±9.1),(107.2±12.1)和(61.9±9.1)U·L^-1;这3组的肝指数分别为(3.40±0.25)×10^-2,(4.50±0.23)×10^-2和(3.90±0.17)×10^-2;这3组的NFκB p65蛋白的相对表达量分别是0.94±0.08,1.27±0.10和0.95±0.07。上述指标:模型组与正常组比较,差异均有统计学意义(P<0.05,P<0.01);实验组与模型组比较,差异均有统计学意义(P<0.05,P<0.01)。结论L-谷氨酰胺可能能够缓解急性酒精肝损伤,其机制可能与谷氨酰胺抑制NF-κB p65的表达有关。Objective To investigate the effect and mechanism of L-glutamine on acute alcohol liver injury and the expression of nuclear factor-kappa B p65(NF-κB p65).Methods Kunming mice were randomly divided into three groups:normal group,model group and experimental group(800 mg·kg^-1·d^-1 L-glutamine),each group had 15 mice.After 1 week of intragastric administration,acute alcoholic liver injury in mice was induced by intragastric administration with 56-degree Hongxing Erguotou.The activities of glutamic-oxaloacetic transaminase(GOT),glutamic-pyruvic transaminase(GPT)were detected by automatic biochemical analyzer.The relative expression(relative gray value)of NF-κB p65 protein was determined of by Western blot.Results The activities of GOT in normal group,model group and experimental group were(113.9±11.4),(203.6±22.1),(158.6±16.8)U·L^-1;the activities of GPT in the three groups were(52.8±9.1),(107.2±12.1),(61.9±9.1)U·L^-1;the liver index in the three groups were(3.40±0.25)×10^-2,(4.50±0.23)×10^-2,(3.90±0.17)×10^-2;the relative expression of NFκB p65 protein in the three groups were 0.94±0.08,1.27±0.10,0.95±0.07.Comparison between model group and normal group,the difference of the factors were significantly(P<0.05,P<0.01);comparison between experimental group and model group,the difference of the factors were significantly(P<0.05,P<0.01).Conclusion L-glutamine can relieve acute alcoholic liver injury,and its mechanism may be inhibit the expression of NF-κB p65 expression.
