摘  要:目的探究亚低温治疗对新生大鼠缺氧缺血性脑病(HIBD)损伤后神经细胞增殖和凋亡的影响。方法将7日龄大鼠45只随机分为对照组、HIBD组和亚低温干预组,每组各15只。对照组为假手术组,即分离左颈总动脉但不结扎,HIBD组分离左颈总动脉并予以结扎,亚低温干预组在左颈总动脉结扎基础上联合亚低温治疗。其中对照组及HIBD组保持肛温36℃~37℃,亚低温组保持肛温32℃~33℃。三组大鼠自10日龄开始均连续腹腔注射5-溴脱氧尿嘧啶核苷(BrdU)用于标记新生细胞。采用免疫组化法检测脑组织海马齿状回颗粒下层BrdU和活化半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)的表达情况,采用苏木素-伊红(HE)染色法计数三组大鼠海马齿状回单位面积神经细胞数并加以比较。结果亚低温干预组BrdU阳性细胞表达明显高于HIBD组,但仍低于对照组(假手术组),差异有统计学意义(P<0.05),亚低温干预组活化Caspase-3表达明显低于HIBD组,但高于对照组,差异有统计学意义(P<0.05)。亚低温组海马齿状回单位面积神经细胞数明显低于对照组(P<0.05),但明显高于HIBD组,差异有统计学意义(P<0.05)。三组大鼠实验开始前体重比较无统计学意义(P>0.05),取脑组织前对照组体重明显重于其余两组,差异有统计学意义(P<0.05),但亚低温组与HIBD组无明显差异(P>0.05)。结论亚低温对大鼠HIBD的海马区神经细胞的增生起促进作用,并减轻其凋亡。Objective To investigate the effect of mild hypothermia on the proliferation and apoptosis of neural cells in neonatal rats with hypoxic-ischemic encephalopathy(HIBD).Methods Forty five 7-days-old rats were randomly divided into control group,HIBD group and mild hypothermia intervention group with 15 rats in each group.The control group was sham operated group,i.e.left common carotid artery was separated but did not ligated,left common carotid artery was separated and ligated in HIBD group,mild hypothermia intervention group was combined with mild hypothermia treatment on the basis of ligation of left common carotid artery.The anal temperature was kept at 36 ℃~37 ℃ in the control group and HIBD group,and the anal temperature was kept at 32 ℃~33 ℃ in the mild hypothermia group.5-bromodeoxyuridine(BrdU)was continuously injected into the abdominal cavity of the three groups of rats since the age of 10 days to mark the renew cells.The expression of BrdU and Caspase-3 in the subgranular layer of dentate gyrus of hippocampus was detected by immunohistochemistry,and the number of nerve cells per unit area of dentate gyrus of hippocampus was counted and compared by hematoxylin eosin(he)staining.Results The amount of BrdU positive cells in mild hypothermia intervention group was significantly higher than that in HIBD group,but still lower than that in control group(sham operation group),the difference was statistically significant(P<0.05).The expression of activated caspase-3 in mild hypothermia intervention group was significantly lower than that in HIBD group,but higher than that in control group,the difference was statistically significant(P<0.05).The number of neurons per unit area of dentate gyrus in mild hypothermia group was significantly lower than that in control group(P<0.05),but significantly higher than that in HIBD group(P<0.05).There was no significant difference in body weight between the three groups before the experiment(P>0.05).The body weight of the control group was significantly heavier than t
