摘  要:为研制鸡Ⅰ群禽腺病毒血清4型(FAdV-4)油乳剂灭活疫苗,试验采用鸡肝癌细胞(LMH)扩增FAdV-4,测定FAdV-4最佳灭活条件和疫苗最佳油水比,研制了FAdV-4油乳剂灭活苗。按《中华人民共和国兽药典》进行疫苗质量、安全性和临床效力检验。结果显示:LMH细胞接种FAdV-4 72h后其毒力最高,TCID50=10-7.78/0.1mL;终浓度0.15%的甲醛溶液37℃24h可灭活FAdV-4;油水比为2∶1的疫苗性状最稳定,质量和安全性检验均合格;疫苗最小免疫剂量为0.4 mL/只;抗体水平在免疫后第2周开始上升,第4周达到最高,而后缓慢下降。对疫苗免疫鸡14 d后注射0.2 mL FAdV-4病毒液(含10-5.78TCID50/0.1 mL),经14 d观察可见鸡全部存活,剖检后无病理变化且攻毒后第3、5、7、10、14天PCR检测均为阴性,临床保护率为100%。研究提示:LMH细胞可有效提高FAdV-4毒价,通过优化FAdV-4灭活条件和疫苗油水比,所研制的疫苗安全性高,保存期长,能有效预防FAdV-4感染,为鸡心包积液综合征防控提供参考。In order to develop an oil emulsion inactivated vaccine against fowl adenovirus group Ⅰ serotype 4(FAdV-4), leghorn male hepatocellular cells(LMH) were used to amplify FAdV-4, the optimal inactivation conditions of FAdV-4 and the optimal oil-water ratio of vaccine were determined, the oil emulsion inactivated vaccine against FAdV-4 was developed. The quality, safety and clinical efficacy of the vaccine were tested according to Veterinary Pharmacopoeia of the People′s Republic of China. The results showed that FAdV-4 infected by LMH reached the highest virulence at 72 h, TCID50=10-7.78/0.1 mL. The formaldehyde solution with a final concentration of 0.15% could inactivate FAdV-4 at 37 ℃ for 24 hours. The vaccine with oil-water ratio of 2∶1 had the most stable traits, quality and safety tests were qualified. The minimum immune dose of the vaccine was 0.4 mL per chicken. The antibody level began to increase in the 2 nd week after immunization, reached the highest in the 4 th week, and gradually declined in the later period. After 14 days of immunization, chickens were injected with 0.2 mL of FAdV-4 virus solution(containing 10-5.78 TCID50/0.1 mL), all the chickens survived 14 days post inoculation, and had no pathological changes after necropsy,PCR tests were negative on the 3 rd, 5 th, 7 th, 10 th, and 14 th days post inoculation,the clinical protection rate was 100%. Study suggested that LMH could effectively increase the virulence of FAdV-4, by optimizing the inactivation conditionsand the oil-water ratio, the developed vaccine had high safety and long protection duration, could effectively prevent FAdV-4 infection and provide reference for the prevention and control of FAdV-4.
