摘 要:柱头在花粉黏附、水合和花粉管生长中具有重要作用,柱头蛋白参与多种生物学过程。为比较亚洲百合与铁炮百合的柱头蛋白质差异,本研究以亚洲百合‘Brunello’和铁炮百合品种‘White Heaven’为试验材料,利用iTRAQ技术分析两种百合的柱头蛋白差异。结果表明亚洲百合和铁炮百合的柱头蛋白总数为3 963,参与的主要生物过程为代谢过程、细胞过程和刺激应答。定量分析结果表明,241个蛋白在2种百合的柱头上差异显著,功能涉及碳水化合物与能量代谢和运输、脂质运输和代谢、蛋白代谢、细胞壁/膜/包膜生物发生、无机离子转运和代谢、信号转导和辅酶转运和代谢等,其中涉及碳水化合物与能量代谢的差异蛋白数量较多。本研究为深入了解百合远缘杂交过程中柱头亲和性差异的分子机制提供技术数据。Stigma plays an important role in pollen adhesion,hydration and pollen tube growth.Stigma proteins are involved in a variety of biological processes.In order to compare the differences of stigma proteins between Asiatic and Longiflorum lilies,iTRAQ based proteomic technique was applied to analyze the protein difference between Asiatic lily cultivar ’Brunello’ and Longiflorum lily cultivar ’White Heaven’.A total of 3 963 proteins were identified and quantified in the stigmas of the two types of lily,and they were mainly involved in metabolic processes,cellular processes and stimuli responses.Quantitative analysis showed that 241 proteins were significantly different from the stigma of the two types of lily and they were related to carbohydrate and energy metabolism,transport,lipid metabolism,protein metabolism,cell wall/membrane/envelope biogenesis,inorganic ion transport,signal transduction,coenzyme transport and metabolism.The proteins expressed more differentially among them that were involved in carbohydrate and energy metabolism.This study provides technical data for understanding the molecular mechanism of affinity differences of stigma in the process of lily hybridization.
