摘  要:谷子是中国北方重要的耐旱杂粮作物。胁迫相关蛋白是一类具有A20和(或) AN1结构域的锌指蛋白,广泛参与植物对非生物胁迫的响应。本研究从‘晋谷20’中克隆得到谷子SiSAP8基因,该基因编码区513 bp,编码170个氨基酸,分子量为18.20 kD,等电点为8.28,包含1个A20和1个AN1结构域。在对该基因序列进行多态性分析,共发现5个SNP位点和1个InDel,其中编码区有1处核苷酸变异位点(G/T)为非同义突变,使得缬氨酸变成了苯丙氨酸。利用第81位的SNP位点(A/G)开发了dCAPS标记,命名为‘SNP-81’,该标记的开发为后期功能鉴定及关联分析提供了依据。Foxtail millet is an important minor crop with drought-tolerance in the north of china.Stress associated proteins(SAPs) are the A20/AN1 zinc-finger proteins which involve in abiotic stress tolerance in plants.In this study,stress associated protein gene SiSAP8 was isolated from foxtail millet variety ’Jingu20’.The full-length sequence of SiSAP8 gene is 513 bp,encoding 170 amino acids.Molecular weight and isoelectric point of SiSAP8 are 18.20 kD and 8.28,respectively.There are A20 domain and AN1 domain in SiSAP8.Five SNPs and one InDel were detected in the sequence.One SNP(G/T) was in the coding region which led to changes of amino acids(Val/Phe).On the basis of the SNP(A/G),a dCAPS marker was developed,named as SNP-81.The dCAPS marker of SNP-81 provided the basis for further function studies and association analysis.
