附子茎段组培快繁体系的建立

摘　　要：为探讨附子丛生芽的诱导、增殖及不定根诱导条件。本研究以附子带腋芽茎段为外植体,以MS和1/2MS为基本培养基,添加不同浓度的6-BA、NAA、TDZ、IBA等植物生长调节剂,观察附子丛生芽的诱导、增殖、生根情况。结果发现,附子茎段经75%乙醇处理30 s后,0.1%Hg Cl2灭菌10 min,污染率为27.78%,存活率可达84.61%。附子第三个腋芽,诱导率为53.34%,死亡外植体少。丛生芽在MS+6-BA 2 mg/L+NAA0.3 mg/L条件时诱导率为86.67%,芽长1.947 cm,植株茎干粗壮。增殖培养时添加TDZ 2 mg/L+NAA0.3 mg/L,增殖系数达到4.029,苗粗壮,叶片浓绿。生根培养基条件为1/MS+IBA 0.5 mg/L时,15 d的生根率可达100%,平均根长0.906 cm,平均根数10.5条,叶色翠绿,生长旺盛。研究表明,最佳的取材部位为第三个腋芽,丛生芽诱导的最佳培养基为MS+6-BA 2 mg/L+NAA 0.3 mg/L,丛生芽增殖培养基中添加TDZ 2 mg/L+NAA 0.3 mg/L的增殖效果最好,适宜的生根培养基为1/2MS+IBA 0.5 mg/L。本研究为附子快繁体系的建立和工厂化育苗提供了理论依据。In order to investigate the induction,proliferation and adventitious root induction conditions of aconite buds.In this study,the aconite bud stems were used as explants,MS and 1/2 MS were used as the basic medium,and different concentrations of 6-BA,NAA,TDZ,IBA and other plant growth regulators were added to observe the induction of aconite shoots,proliferation and rooting.The result showed that after treatment with 75% ethanol for 30 s,0.1% Hg Cl2 was sterilized for 10 min,the contamination rate was 27.78%,and the survival rate was84.61%.The third axillary bud of aconite had an induction rate of 53.34% and less dead explants.The induction rate of cluster buds in the condition of MS+6-BA 2 mg/L+NAA 0.3 mg/L was 86.67%,the bud length was 1.947 cm,and the stems of the plants were thick.During the proliferation culture,TDZ 2 mg/L+NAA 0.3 mg/L was added,and the proliferation coefficient reached 4.029.The seedlings were thick and the leaves were dark green.When the rooting medium condition is 1/2 MS+IBA 0.5 mg/L,the rooting rate can reach 100% in 15 days,the average root length is 0.906 cm,the average root number is 10.5,the leaf color is green and the growth is strong.The research found that,the best material is the third axillary bud.The best medium for shoot bud induction is MS+6-BA 2 mg/L+NAA 0.3 mg/L.TDZ is added to the shoot bud proliferation medium.2 mg/L +NAA 0.3 mg/L had the best proliferation effect,and the suitable rooting medium was 1/2 MS+IBA 0.5 mg/L.This study provides a theoretical basis for the establishment of aconite rapid propagation system and factory breeding.
