摘 要:为优化新兴的、现存量稀少的木本花卉红冠桉(Corymbia ptychocarpa)腋芽快繁体系。本研究以15年生母树无菌茎段为外植体,利用茎段萌发腋芽的方式,筛选多腋芽启动方法;通过正交试验设计方法对丛生芽形成、芽增殖和生根诱导等培养基进行优化,研究不同栽培基质对幼苗移栽驯化的影响。结果表明,经预处理的茎段以MS+0.5 mg/L 6-BA为腋芽启动培养基较适宜,萌发率达97.1%,平均发芽系数3.6;不同浓度6-BA与ZT组合试验表明,在MS+0.5 mg/L ZT+6-BA 3.0 mg/L培养基中,丛生芽最大体积达2.51 cm3;不同激素配比试验表明,MS+0.8 mg/L 6-BA+0.03 mg/L ZT+0.04 mg/L IBA培养基对促进芽增殖、生长最佳,增殖系数6.2±1.34,嫩茎平均高度2.60 cm;生根试验中,提高MS培养基中铁盐浓度,生根率可达96.30%,平均根条数6条/株;组培苗移栽至当地土(红壤)基质中,平均成活率达85.4%。本研究建立了一套完整的红冠桉快繁体系,为其工厂化育苗提供系统的组培技术及遗传改良提供科学依据。In order to optimize the rapid propagation system of axillary buds of Corymbia ptychocarpa which was a burgeoning and rare of existing woody flower.As sterile stems of 15 years old Corymbia ptychocarpa plant as the explants,screening the method of multiple axillary buds initiation by germinating axillary buds of stems.Optimizing the medium of multi-buds formation,shoots proliferation and rooting by the orthogonal test design;searching the different substrates influence on the survival rate of seedlings transplantation.The results showed the better medium was MS+0.5 mg/L 6-BA,when pretreatment stems were germinated axillary buds in it,and the germination rate was 97.1%,the germination coefficient was 3.6;The test of different concentration 6-BA and ZT combination showed the biggest volume of cluster buds was 2.51 cm3 in the medium of MS+0.5 mg/L ZT+6-BA 3.0 mg/L.The test of different phyto-hormone ratio suggested the proliferation coefficient and the height of shoots were 6.2±1.34 and 2.6 cm respectively in the medium of MS+0.8 mg/L 6-BA+0.03 mg/L ZT+0.04 mg/L IBA which was the best for promoting shoot proliferation and growth.The test of rooting indicated raising the concentration of EDTA-Fe solutions in MS medium,the rooting rate and the number of per shoot were 96.30% and 6.0 respectively.The survival rate of tissue culture plantlet was 85.4%,when they were transplanted into the substrate of the garden soil(red soil).A complete set of rapid propagation system of Corymbia ptychocarpa has been established and supplied the systematic technique of tissue culture for its industrialized breeding and the scientific foundation for its genetic improvement.
