摘 要:本研究基于SRAP标记和ITS序列对11份西番莲种质进行遗传多样性分析。结果表明,SRAP标记分析时从70对引物组合中筛选出9对重复性好,条带清晰的引物组合,共扩增出110个条带,平均每组引物可扩增12.22个条带,多态性条带占84.93%;UPGAM聚类可以将11个西番莲样品分为5个类群,第一类:‘哥伦比亚热情果’,‘巨无霸’;第二类:‘黄金果’,‘台农1号’,‘满天星’和‘天王星’;第三类:‘蓝香’和‘瑞香’;第四类:‘玛格丽特’;第五类:‘龙珠果’。ITS序列分析时,11个西番莲样品ITS序列长度为667 bp,变异位点为162个,单倍型10个,单倍型多样性0.982,核苷酸多样性0.071。采用NJ法对11个样品构建系统进化树可分为4大支,‘哥伦比亚热情果’,‘巨无霸’为一支;‘蓝香’,‘瑞香’和‘玛格丽特’为一支;‘金霸’,‘黄金果’,‘台农1号’,‘满天星’和‘天王星’为一支。通过两种方法分析均能很好地将11个西番莲样品区分开来,结果基本一致,且与其属种相吻合,说明SRAP标记和ITS序列对西番莲进行分类是可行的。因此,西番莲遗传多样性能更好地开发和利用种质资源,为西番莲育种与品种鉴定提供科学依据。The genetic diversity of 11 Passiflora germplasm was analyzed based on SRAP and ITS sequences.The results showed that 9 pairs of primers with good repeatability and clear bands were selected from 70 pairs of primers by SRAP marker analysis,and the number of bands of the 9 pairs of primers is 110,with an average of12.22,with polymorphism bands accounting for 84.93%.UPGAM clustering can divide 11 Passiflora into 5 groups,The first:’Colombian passion fruit’,’Juwuba’;the second,’Golden fruit’,’Tainong 1’,’Mantianxing’ and ’Tianwangxing’;the third:’Lanxiang’ and ’Ruixiang’;the fourth:’Margaret’;the fifth:’Longzhuguo’.In the analysis of ITS sequence,the length of ITS sequence of 11 passion flower samples was 667 bp,162 variation sites,haplotype is 10,haplotype diversity was 0.982,nucleotide diversity was 0.071,respectively.The phylogenetic tree constructed by NJ method on 11 samples can be divided into four branches,’Columbia passion fruit’ and ’Juwuba’,’Lanxiang’,’Ruixiang’ and ’Margaret’,’Jinba’,’Golden fruit’,’Tainong 1’,’Mantianxing’ and ’Tianwangxing’ as a branch.The two methods can be used to distinguish 11 Passiflora samples,and the results are basically the same,which is consistent with their genus and species,indicating that it is feasible to classify passion flower with SRAP marker and ITS sequence.Therefore,Passiflora genetic diversity can develop and utilize germplasm resources better,and provide scientific basis for Passiflora breeding and variety identification.
