摘  要:为研究重金属胁迫对秋茄幼苗抗氧化系统的影响,砂培红树植物秋茄幼苗一个月,研究不同重金属浓度(Cu^2+:0、3、15、30、45 mg·L^–1,Pb^2+:0、1、5、10、15 mg·L^–1)和不同实验时间(0、3、7、14、28 d)对叶片的影响。测量的生理指标包括:可溶性蛋白、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和过氧化物酶(POD),实验条件下两种重金属对各项参数的影响一致。结果表明:在特定浓度重金属处理下(Cu2+:0、3、15、30、45 mg·L^–1,Pb^2+:0、1、5、10、15 mg·L^–1),可溶性蛋白含量随着时间的增加基本呈先升后降趋势;SOD活性呈降低趋势;CAT活性最初受到明显促进,后促进作用减弱;POD活性变化规律并不一致。在相同处理时间下,可溶性蛋白含量随Cu2+浓度的增加而降低,在Pb^2+处理下有所波动;SOD活性在各浓度条件下均降低;CAT活性基本呈降低趋势;POD活性呈先升后降的趋势。通过Pearson相关性分析和PCA分析表明,可溶性蛋白含量与SOD、POD、CAT三种酶活性呈显著负相关关系,相关系数分别为–0.401,–0.722,–0.521;而SOD、POD和CAT活性则表现为显著正相关关系,SOD与POD的相关系数为0.359,SOD与CAT的相关系数为0.384,POD与CAT的相关系数为0.485,说明三种抗氧化酶协同作用形成一条抗氧化链抵御重金属胁迫。Mangrove seedlings of Kandelia obovata were cultured in sands for one month.The effects on leaves were studied under different concentration of the heavy metal stress(Cu^2+:0,3,15,30,45 mg·L^−1,Pb^2+:0,1,5,10,15 mg·L^−1)and the different experimental times(0,3,7,14,28 days).The physiological indicators included soluble protein,superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD). Under experimental conditions, the efffects of the two heavy metalson parameters were consistent. At the same heavy metal concentration, the results showed as follows: (1) the soluble proteincontent basically increased first, and then decreased with the increase of the experimental time;(2) the SOD activity showeda decreasing trend, and CAT activity was significantly promoted at first, and then the promotion effect was weakened;(3) thechanges of the POD activity were not consistent. At the same treatment time, the soluble protein content decreased with theincrease of Cu^2+ concentration, and fluctuated with the increase of Pb^2+ concentration. The SOD activity decreased at allconcentrations, the CAT activity showed a decreasing trend, the POD activity increased first, and then decreased. ThePearson analysis and PCA analysis showed that soluble protein content had significantly negative correlation with theactivity of the SOD, POD, CAT. The correlation coefficients were -0.401, -0.722, -0.521;while SOD, POD and CAT activityhad significantly positive correlation. The correlation coefficients between SOD and POD was 0.359, the correlationcoefficients between SOD and CAT was 0.384, and the correlation coefficients between CAT and POD was 0.485. This resultindicated that these three antioxidant enzymes formed an antioxidant chain to resist the stress of heavy metals.
