摘 要:利用响应面法优化芦丁的最佳荧光检测条件。以芦丁浓度、硝酸铝用量、醋酸-醋酸钠缓冲溶液的pH等作为考察因素,以荧光强度作为考察指标,在单因素试验基础上,通过Design-Expert.V 8.0.6.1软件进行三因素三水平试验设计,并对设计的试验结果进行响应面回归模型分析,获得二次多项回归方程。优化后的芦丁荧光检测条件为:芦丁质量浓度37.78 mg/L,0.5%硝酸铝体积2.76 mL,缓冲溶液pH 5.61,其中,缓冲溶液的pH对芦丁的荧光强度影响最显著,芦丁质量浓度为不显著影响因素。The fluorescence detection condition of rutin was optimized by response surface methodology.Taking rutin concentration,aluminium nitrate dosage and pH of acetic acid-sodium acetate buffer as factors,fluorescence intensity as index,on the basis of single factor test,three-factor and three-level test design was carried out by Design-Expert.V 8.0.6.1 software.The test results were analyzed and the quadratic polynomial regression equation was obtained by response surface regression model.The optimized fluorescence detection condition was that concentration of rutin 37.78 mg/L,the volume of 0.5%aluminium nitrate 2.76 mL and the pH 5.61,and the pH of buffer solution had the most significant effect on the fluorescence intensity of rutin,but the concentration of rutin was not significant.
