摘 要:目的考察配伍对麻黄-桂枝药对抗炎作用的影响,并探讨其抗炎机制。方法采用醋酸致腹腔毛细血管通透性增高实验观察麻黄、桂枝及麻黄-桂枝药对的抗炎作用;采用角叉菜胶诱导大鼠胸膜炎实验考察麻黄-桂枝药对的抗炎作用,并通过测定胸腔渗出液的体积、白细胞计数、PGE2、TNF-α、IL-1β的含量及肺部iNOS、ICAM-1免疫组化检测,探讨作用机制。结果麻黄、桂枝及麻黄-桂枝药对不同配比对醋酸致毛细血管通透性的增高具有一定的抑制作用,并呈现一定的量效关系,配伍后抑制率显著升高。麻黄-桂枝药对可以减少胸腔液渗出、白细胞数量及肺损伤;并能降低胸腔炎性渗出液中PGE2、TNF-α、IL-1β的含量及肺部iNOS、ICAM-1的表达。结论麻黄-桂枝药对具有明显的抗炎作用,二者配伍后作用优于单味药,抗炎机制与抑制致炎介质PGE2、细胞因子TNF-α、IL-1β的生成及抑制iNOS、ICAM-1的表达有关。Objective To investigate the anti-inflammatory and mechanism of Herba Ephedrae-Ramulus Cinnamomi(Eph-RC)herb pair with different compatibility.Methods The test of acetic acid-induced peritoneal permeability accentuation was employed for study on the anti-inflammatory effect of Herba Ephedrae(Eph),Ramulus Cinnamomi(RC)and Herba Ephedrae-Ramulus Cinnamomi(Eph-RC)herb pair.Carrageenan-induced pleurisy model of rats was carried out to observe the anti-inflammatory activity.The anti-inflammatory mechanisms were investigated by detecting exudate volume,white blood cell count and the contents of prostaglandins(PGE2),tumor necrosis factor-alpha(TNF-α),interleukin-lβ(IL-1β)in exudates.In addition,pathological sections,iNOS and ICAM-1 immunohistochemical detection in the lungs were also performed.Results Eph group,RC group and Eph-RC of different ratio group can significantly inhibit acetic acid-induced peritoneal permeability increase in mice,and presented a dose-response relationship.The inhibitory effect increased significantly after compatibility.Eph-RC herb pair can significantly inhibit carrageenan-induced pleural effusion,leukocytes increase and lung injury in rats.The content of PGE2,TNF-αand IL-1βin exudate was significantly decreased.Immunohistochemical detection results showed that Eph-RC can significantly inhibit the expression of iNOS and ICAM-1.Conclusion Eph-RC herb pair has obvious inhibitory effect on inflammation,and the combination of Eph and RC is more effective than single use.The anti-inflammatory mechanism of Eph-RC herb pair is related to inhibiting generation of PGE2,TNF-α,IL-1βand the expression of iNOS and ICAM-1.
