活血清解汤对急性胰腺炎大鼠的调节作用及相关机制研究

摘　　要：目的探讨活血清解汤对急性胰腺炎大鼠的调节作用及其相关机制。方法将18只SD大鼠随机分成空白对照组(C组)、模型组(M组)和活血清解汤治疗组(T组),每组6只。用20%L-精氨酸溶液腹腔注射大鼠,建立急性胰腺炎大鼠模型。采用HE染色观察胰腺组织的形态,采用qPCR、Western blot、免疫组化和ELISA法检测HO-1、IL-1β、IL-6、TNF-α、GSTM3和RyR的表达情况。结果与C组相比,M组胰腺组织损伤较严重,用活血清解汤处理后,其严重程度显著下降。与M组相比,在T组中HO-1、RyR水平显著增加,IL-1β、IL-6、TNF-α表达水平显著降低,而GSTM3的表达无显著变化。结论活血清解汤对急性胰腺炎具有良好的保护作用,其可能通过上调HO-1的表达,抑制促炎因子的产生及调节氧化应激反应来发挥作用。Objective To investigate the regulatory effect of Huoxue Qingjie decoction on rats with acute pancreatitis and its related mechanism.Methods Totally 18 SD rats were randomly divided into blank control group(group C),model group(group M)and Huoxue Qingjie decoction group(group T),6 rats in each group.Acute pancreatitis model was established by intraperitoneal injection of 20%L-arginine solution into rats.The morphology of pancreatic tissue was observed by HE staining,and the expressions of HO-1,IL-1β,IL-6,TNF-α,GSTM3 and RyR were detected by qPCR,Western blot,immunohistochemistry and ELISA.Results Compared with group C,the pancreatic tissue injury in group M was more serious,which was relieved after treatment of Huoxue Qingjie decoction.In addition,compared to group M,HO-1 and RyR levels in group T were significantly increased,while the levels of IL-1β,IL-6 and TNF-αwere significantly decreased;GSTM3 expression was not significantly changed.Conclusion Huoxue Qingjie decoction has a good protective effect on acute pancreatitis by up-regulating the expression of HO-1,inhibiting the production of pro-inflammatory factors and regulating the oxidative stress response.
