摘  要:采用腺嘌呤造模的大鼠慢性尿酸性肾病为模型,探讨鸡矢藤环烯醚萜苷类化合物(IGsP)在治疗尿酸性肾病中的应用。将SD大鼠随机分为正常对照组、模型组,IGsP低、中、高剂量(50、100、200 mg/kg)组和阳性药别嘌醇(20 mg/kg)组、苯溴马隆(20 mg/kg)组。各模型和给药组大鼠按0.4%腺嘌呤饲料给药的方法造模,造模2周后开始给药,为期2周,治疗期间检测大鼠血清内尿酸(UA)、尿素氮(BUN)、肌酐(Cre)含量,试验结束后大鼠处死取肾脏称重,计算脏器指数后切片染色,观察肾脏组织的病理学变化。结果显示与模型组相比,不同剂量的IGsP均可降低慢性尿酸性肾病大鼠体内的尿酸(UA)、尿素氮(BUN)、肌酐(Cre)水平,并呈现一定的时间和剂量依赖性,且高剂量组的降尿酸效果与阳性对照药相似;通过观察大鼠的脏器指数和肾脏切片发现,不同剂量的IGsP均可降低尿酸性肾病大鼠肾脏指数,对大鼠肾脏病理损伤也有部分改善作用。结果显示IGsP对腺嘌呤诱导的大鼠尿酸性肾病具有一定的治疗效果。To Study on the application of iridiod glycosides from Paederia scandens L.on uric acid nephropathy in rats induced by adenine.SD rats were randomly divided into 7 groups:control group,model group,positive drug allopurinol(20 mg/kg)group and benzbromarone(20 mg/kg)group,and low,medium,high dose group of iridiod glycosides Paederia scandens L.(50,100,200 mg/kg).The rats in model and administration groups were given 0.4%adenine feed to establish the model for four weeks.The drugs were administered for 2 weeks(at last two weeks of the molding).The serum of rats were detected during the treatment period.Meanwhile the contents of uric acid(UA),urea nitrogen(BUN)and creatinine(Cre)were measured.At the end of the experiment,the rats were executed and their kidneys were weighed to calculate the organ index,and the pathological changes of kidney tissue were observed with H&E and MASSON staining.Compared with the model group,different doses of iridiod glycosides from Paederia scandens L.could reduce the serum levels of uric acid(UA),urea nitrogen(BUN)and creatinine(Cre)of rats with uric acid nephropathy in a time-and dose-dependent manner,and the effect of reducing uric acid in the high dose group was similar to that in the positive control group.The same dosage of iridiod glycosides from Paederia scandens L.could reduce the renal index of rats with uric acid nephropathy and improve the pathological damage of kidneys of the rats.The iridiod glycosides from Paederia scandens L.has a certain therapeutic effect on adenine-induced uric acid nephropathy in rats.
