摘 要:目的观察温阳平喘方对哮喘脾阳虚证小鼠血清中胃泌素(Gas)、肿瘤坏死因子-α(TNF-α)及肺泡灌洗液(BALF)中免疫球蛋白E(IgE)、神经生长因子(NGF)和酪氨酸激酶A(TrkA)水平的影响。方法将60只BALB/c雌性小鼠按体质量随机分为6组,即空白组,模型组,温阳平喘方低、中、高剂量组(分别简称低、中、高剂量组),地塞米松组(简称地米组)。除空白组外,其余各组小鼠连续10 d采用饮食不节、过度劳倦和苦寒泄下多种因素进行脾阳虚证造模。造模成功后,使用卵清白蛋白(OVA)致敏和雾化吸入激发的方法复制哮喘模型。采用ELISA法检测血清中Gas、TNF-α水平及BALF中IgE、NGF和TrkA水平。结果与空白组比较,模型组Gas水平明显降低,TNF-α、IgE、NGF、TrkA水平均明显升高(P<0.01)。与模型组比较,中、高剂量组及地米组Gas水平升高(P<0.01);低、中、高剂量组及地米组TNF-α及IgE、NGF、TrkA水平均明显降低(P<0.05或P<0.01)。与地米组比较,中、低剂量组TNF-α及IgE、NGF、TrkA水平差异有统计学意义(P<0.05或P<0.01)。高、中、低剂量组组间比较,高剂量组的疗效优于其他2组(P<0.05)。结论温阳平喘方能够明显升高哮喘脾阳虚证小鼠血清中Gas水平,降低IgE、TNF-α、NGF及TrkA水平,改善脾胃运化功能,降低炎性细胞因子水平,从而减轻炎症反应。Objective To observe the effects of Wenyang Pingchuan Fang(温阳平喘方,WPF)on levels of gastrin(Gas),tumor necrosis factor-α(TNF-α),immunoglobulin E(IgE),nerve growth factor(NGF)and tyrosine kinase A(TrkA)in bronchoalveolar lavage fluid(BALF)in the mice model of asthma with spleen yang deficiency syndrome.Methods Sixty BALB/c female mice were randomly divided into 6 groups according to body mass,namely blank group,model group,WPF low,medium and high dose group and dexamethasone group.Except the blank group,the mice in the other groups were given multi-factor involving improper diet,overstrain,diarrhea with bitter-cold herbs for consecutive 10 days to establish the models with deficiency of spleen yang syndrome.After successfully establishing the models,we replicated asthma model by using the method of ovalbumin(OVA)sensitization and aerosol inhalation challenge.Enzyme linked immunosorbent assay(ELISA)method was used to detect the levels of Gas and TNF-αin serum and the levels of IgE,NGF and TrkA in BALF.Results Compared with those of the blank group,the level of Gas decreased significantly,while the levels of TNF-α,IgE,NGF and TrkA in the model group increased significantly(P<0.01).Compared with those of the model group,the level of Gas increased significantly in WPF medium,high dose group and dexamethasone group(P<0.01),while the levels of TNF-α,IgE,NGF and TrkA significantly decreased in WPF low,medium and high doze group and dexamethasone group(P<0.05 or P<0.01).Compared with those of the dexamethasone group,the difference was statistically significant in the levels of TNF-α,IgE,NGF and TrkA of WPF low,medium dose group(P<0.05 or P<0.01).Compared among WPF high,medium and low dose group,the efficacy of the high dose group was better than that of the other two groups(P<0.05).Conclusion WPF can significantly increase Gas level,decrease IgE,TNF-α,NGF and TrkA levels,improve spleen and stomach transport function,reduce inflammatory cytokines level in the mice model of asthma with spleen yang deficiency
