摘  要:目的观察加减温经汤对妇科寒凝血瘀证大鼠Rho/ROCK信号通路相关因子表达的影响,探讨其调节微血管的作用机制。方法实验大鼠随机分为空白组、模型组、BQ123组和温经汤组,冰水浴法建立妇科寒凝血瘀证大鼠模型,检测大鼠全血血液流变学相关指标,ELISA检测血清和卵巢组织内皮素(ET)-1和一氧化氮(NO)含量,RT-qPCR和Western blot分别检测卵巢组织Rho/ROCK信号通路关键因子RhoA、ROCK1、MLCK mRNA和蛋白的表达。结果与空白组比较,模型组大鼠全血血液流变学相关指标均明显升高(P<0.01,P<0.05),血清和卵巢组织ET-1含量明显升高,NO含量明显降低(P<0.01),卵巢组织RhoA、ROCK1、MLCK mRNA和蛋白表达明显升高(P<0.01);与模型组比较,BQ123组和温经汤组大鼠血清和卵巢组织ET-1含量明显降低,NO含量明显升高(P<0.01,P<0.05),卵巢组织RhoA、ROCK1、MLCK mRNA和蛋白表达明显降低(P<0.01)。结论加减温经汤通过抑制Rho/ROCK信号通路相关因子的过度激活,改善卵巢子宫组织微血管调节的失衡,从而治疗妇科寒凝血瘀证。Objective To explore the effects of modified Wenjing Decoction on Rho/ROCK signaling pathway related factors expression in rats with gynecological cold coagulation and blood stasis syndrome;To explore the mechanism of its role in regulating microvessels.Methods The experimental rats were divided into blank group,model group,BQ123 group and Wenjing Decoction group.Rat models of gynecological cold coagulation and blood stasis syndrome were established by ice-water bath method.Whole blood rheology was measured in each group.ET-1 and NO in serum and ovarian tissue were detected by ELISA.RT-qPCR and Western blot were used to detect the expression of RhoA,ROCK1,MLCK mRNA and protein,which were the key factors of Rho/ROCK signaling pathway in ovarian tissue.Results Compared with the blank group,the whole blood rheological indexes in the model group increased(P<0.01,P<0.05),the ET-1 content in serum and ovarian tissue increased,NO content decreased significantly(P<0.01),and the RhoA,ROCK1,MLCK mRNA and protein expression in ovarian tissue increased significantly(P<0.01).Compared with the model group,the ET-1 content in serum and ovarian tissue decreased significantly in BQ123 and Wenjing Decoction groups,NO content increased significantly(P<0.01,P<0.05),and RhoA,ROCK1,MLCK mRNA and protein in ovarian tissue significantly decrease(P<0.01).Conclusion Modified Wenjing Decoction can improve the imbalance of microvascular regulation in ovary and uterus,and treat gynecological cold coagulation and blood stasis syndrome by inhibiting the over-activation of Rho/ROCK signaling pathway related factors.
