摘  要:目的:构建胡桃楸根指纹图谱,并采用超高效液相色谱-三重四级杆串联质谱法(UPLC-Q-TOF-MS/MS)分析鉴定胡桃楸根中的化学成分。方法:采用高效液相色谱法(HPLC)建立胡桃楸根HPLC指纹图谱,通过相似度评价和主成分分析对23批胡桃楸根药材进行质量分析。并采用UPLC-Q-TOF-MS/MS,根据化合物的精确分子量和碎片信息,结合文献数据与课题组前期的研究结果鉴定胡桃楸根中的化学成分。结果:23批胡桃楸根样品中确定了18个共有峰,其相似度为0.731~0.986。主成分分析表明,12个成分对不同批次胡桃楸根指纹图谱的差异贡献率较大。在胡桃楸根样品中共鉴定了88个化学成分,其中包括16个共有成分和11个对胡桃楸根指纹图谱差异贡献率较大的成分。结论:胡桃楸根HPLC指纹图谱的建立以及化学成分的鉴定,为胡桃楸根的质量评价奠定了基础。Objective:To establish the fingerprint of Juglans mandshurica root and identify the chemical constituents in Juglans mandshurica root by UHPLC-Q-TOF-MS/MS.Methods:HPLC fingerprint of Juglans mandshurica root was established by HPLC method.The quality of 23 batches of Juglans mandshurica root was analyzed by similarity evaluation and principal component analysis.In addition,UHPLC-Q-TOF-MS/MS was used to identify the chemical constituents in Juglans mandshurica root.Combining with the previous research results of our research group,the chemical constituents in Juglans mandshurica root were identified by comparing the accurate molecular weight,retention time and secondary mass spectrum characteristics of each ion peak with those of reference substance or those reported in the literature.Results:18 common peaks were identified in 23 batches of Juglans mandshurica root samples,and their similarity ranged from 0.731 to 0.986.According to PCA,12 constituents that play a significant role in distinguishing the differences of Juglans mandshurica root fingerprints were ascertained.A total of 88 chemical components were identified in the roots of Juglans mandshurica,including 16 common constituents and 11 chemical constituents that played a significant role in distinguishing the differences of Juglans mandshurica root fingerprints.Conclusion:The establishment of HPLC fingerprint of Juglans mandshurica root combined with the identification of the chemical constituents of the root by UHPLC-Q-TOF-MS/MS method has laid a foundation for the quality evaluation of J uglans mandshurica root.
