摘  要:目的:应用代谢组学技术对重症脓毒症患者血液变化进行特征分析,以期获得能够表征脓毒症发展变化的新生物标志物。方法:收集重症脓毒症患者血液样本8例,健康志愿者血浆样本7例;应用固相萃取技术处理样本后,进行高效液相色谱串联四级杆飞行时间质谱分析,结合主成分分析和偏最小二乘法分析法,探讨患者组和正常组之间主要代谢产物的差异。结果:患者组和正常组代谢轮廓分析得到了很好的区分,发现了8种潜在的生物标志物,其中6种为花生四烯酸类物质。结论:通过代谢组学分析可以初步判断花生四烯酸类物质在重症脓毒症疾病发生过程中存在一定代谢变化,具有成为表征疾病发展变化的生物标志物的潜在研究价值。Objective To obtain new biomarkers of severe sepsis using metabolomics technique. Methods Plasma samples of 8 patients and 7 health volunteers were collected. After solid phase extraction, samples were assayed by LC-Q-TOF in negative mode. Data was analyzed by principal analysis(PCA) and partial least squares discriminant analysis(PLS-DA) to discover compounds with significant difference. Results The patient group’s metabolites have difference with the control group. And we found 8 potential biomarkers in human plasma and 6 of them were associated with Arachidonic acid. Conclusion We developed an effective pathway to find biological biomarkers in sever sepsis patient’ s plasma and confirmed that Arachidonic acid and several other fatty acids had potential value in revealing pathogenesis of severe sepsis.
