摘  要:目的探讨电针(electroacupuncture,EA)刺激对大鼠条件性位置回避(conditioned place avoidance,CPA)行为反应的影响及其可能的作用机制。方法免疫荧光染色法检测正常大鼠前扣带皮层(anterior cingulate cortex,ACC)脑区组织中μ-阿片受体和NR1的共表达情况。于大鼠ACC脑区内分别预先微量注射μ-阿片受体拮抗剂CTOP及0.9%氯化钠(NS)后,经左脚掌皮下注射完全弗氏佐剂(complete Freund adjuvant,CFA)诱导产生慢性炎性疼痛,并与特定环境进行匹配,建立CPA模型,再进行环跳穴位电针(electroacupuncture,EA)刺激(即CTOP+CFA+EA组和NS+CFA+EA组),同时设假EA刺激组(EA不通电,即CTOP+CFA+shamEA组和NS+CFA+shamEA组)及对照组(ACC及脚掌均注射NS,EA不通电,即NS+NS+shamEA组),记录CPA试验的第1、3天各组大鼠在痛侧匹配室停留时间,计算回避分数。将各组大鼠放置50℃热板上,检测EA刺激对大鼠热痛行为反应的影响。Western blot法检测各组大鼠ACC脑区组织中μ-阿片受体及磷酸化NR1(p-NR1)的表达水平。结果μ-阿片受体与NR1在正常大鼠ACC脑区同一个神经元上共表达。NS+CFA+shamEA组大鼠第3天在痛侧停留时间比第1天显著缩短(P<0.001),回避分数比NS+NS+shamEA组显著增大(P<0.001);NS+CFA+EA组大鼠第3天在痛侧停留时间与第1天相比,差异无统计学意义(P>0.05),回避分数比NS+CFA+shamEA显著减小(P<0.001)。EA刺激不影响大鼠的热痛行为反应。与NS+CFA+shamEA组比较,NS+CFA+EA组大鼠ACC脑区μ-阿片受体表达量明显增高(P<0.001),p-NR1蛋白表达量明显降低(P<0.001)。CTOP可明显逆转EA刺激对CFA诱导CPA反应及ACC脑区μ-阿片受体、p-NR1的表达水平的影响(P<0.001)。结论大鼠CPA反应可能是由CFA诱导ACC脑区p-NR1水平增高导致的,而EA刺激可通过激活μ-阿片受体来减弱该反应。Objective To observe the effect of electroacupuncture(EA)on conditional position avoidance(CPA)behavior response in rats as well as the possible mechanism.Methods The co-expression ofμ-opioid receptor and NR1 in anterior cingulated cortex(ACC)of normal rats was determined by immunofluorescent staining.Rats were randomly divided into five groups.The rats in CTOP+CFA+EA and NS+CFA+EA groups received the microinjection withμ-opioid receptor antagonist CTOP and 0.9 sodium chloride(normal saline,NS)in the ACC respectively,followed by subcutaneous injection with complete Freund adjuvant(CFA)in the left hind paw to induce chronic inflammatory pain,and to establish a CPA model based on the matching to the specified environment,while the model rats received electroacupuncture(EA)stimulation in Huantiao acupoint.However,the rats in CTP+CFA+shamEA and NS+CFA+shamEA groups received sham EA stimulation(without current passing).The rats in control(NS+NS+sham EA)group were injected with NS in both ACC and paw and received sham EA stimulation.The residence time of rats in conditional matching chamber on days 1(one day before EA stimulation)and 3(one day after EA stimulation)of CPA test were measured to calculated the score for avoidance.The rats in various groups were put on the 50℃heat plate to evaluate the effect of EA stimulation on heat pain behavior response.The expression levels ofμ-opioid receptor and phosphorylated NR1(pNR1)in ACC of rats in various groups were determined by Western blot.Results Theμ-opioid receptor and NR1 were co-expressed on the same neuron in ACC of normal rats.The residence time of rats in NS+CFA+shamEA group in conditional matching chamber on day 3 of CPA test was significantly shorter than that on day 1(P<0.001),while the score for avoidance was significantly higher than that in NS+NS+shamEA group(P<0.001).However,the residence time of rats in NS+CFA+EA group in conditional matching chamber on day 3 of CPA test showed no significant difference with that on day 1(P>0.05),which the score for a
