摘 要:为给云南省自育茄子品种‘云茄3号’的制种和大面积推广应用提供技术支持,研究利用SSR分子标记技术对‘云茄3号’的亲本(母本E26×父本E268)进行特异引物筛选,并将筛选出的呈共显性的多态性引物在市场常见茄子品种中进行复筛,应用最终筛选出的标记引物对‘云茄3号’单株进行纯度鉴定,对比田间植株纯度检测结果,验证SSR分子标记种子纯度鉴定的有效性.研究结果表明,从66对已报道的备筛SSR引物中,筛选出2对引物(SM17和SM29)在亲本中呈互补带型的多态性引物,2对引物均能清晰地将‘云茄3号’与其它6个茄子杂交品种区分开来.纯度鉴定结果显示,SSR分子标记检测与田间植株纯度鉴定结果一致,均达到了99%.以上结果表明,在检测种子纯度方面,SSR分子标记与传统的田间种植观察相比有着简单、高效的特点,可作为分子标记辅助手段,加快茄子优良品种的选育和推广.In order to provide a technical support for seed production and large-scale application of eggplant cultivar‘Yunqie No.3’in Yunnan Province,SSR molecular marker technology was used to screen the specific primers for the parents of‘Yunqie No.3’(E26×E268),then the codominant polymorphic primers were screened in common eggplant cultivars in the market,and the final selected pairs of marker primers were applied.The purity of single plant of‘Yunqie No.3’was identified,and the results of plant purity test in the field were compared to verify the validity of SSR molecular marker for purity identification.The results showed that two pairs of primers(SM17 and SM29)were selected from 66 pairs of reported screening primers,which showed complementary and polymorphism band types in their parents.They could clearly distinguish‘Yunqie No.3’from the other six eggplant hybrids.The results of purity identification showed that detection of SSR molecular marker was the same as that of plant purity identification in the field,reaching 99%.The above results showed that SSR molecular markers were simple and efficient in detecting seed purity compared with the traditional field observation,and could be used as an assistant method to accelerate the breeding and promotion of eggplant varieties.
