摘  要:目的研究燃煤型氟中毒对雌性大鼠雌激素的影响在骨代谢中的作用。方法50只SD雌性大鼠按体重随机分为对照组、低氟组、中氟组、高氟组、去卵巢组,每组10只,其中低、中、高氟组分别食用含氟量为25、45、110 mg/kg的病区玉米饲料150 d。检测氟斑牙发生率,采用显微CT技术检测大鼠骨矿物质密度(BMD),以放射免疫法检测大鼠血清雌二醇(E2)含量,以酶联免疫吸附法(ELISA)检测大鼠血清骨代谢标志物。结果成功建立燃煤型氟中毒雌性大鼠模型。中、高氟组大鼠BMD均低于对照组,低氟组BMD高于对照组,差异有统计学意义(P<0.05);中、高氟组E2低于对照组,差异有统计学意义(P<0.05);各染氟组骨钙素(BGP)、骨碱性磷酸酶(BALP)、胰岛素生长因子-1(IGF-1)、胰岛素生长因子结合蛋白5(IGFBP5)均低于对照组,差异有统计学意义(P<0.05);各染氟组抗酒石酸酸性磷酸酶5b(TRACP-5b)、胰岛素生长因结合蛋白4(IGFBP4)均高于对照组,差异有统计学意义(P<0.05)。结论燃煤型氟中毒雌性大鼠对E2分泌功能的损伤可能加重了氟骨症发生,IGF家族可能介导该过程。Objective To understand the effects of chronic coal-burning fluoride exposure-induced estrogen change on bone metabolism of female rats.Methods A total of 50 female SD rats were randomly divided into control group,low-fluoride group,moderate-fluoride group,high-fluoride group,and ovariectomized group according to body weight.The low-,moderateand high-fluoride groups were fed with the corn fodder from endemic fluorosis area which containing fluoride of 25,45 and110 mg/kg respectively,for 150 consecutive days.The dental fluorosis was examined,and BMD was detected by micro-CT technology,serum E2 content were detected by RIA,and rat bone metabolism markers were determined by ELISA.Results Female rats model of coal-burning fluorosis was successfully established.The BMD in moderate-and high-fluoride groups were significantly lower compared with the control group(P<0.05).BMD in low-fluoride group was higher than that in the control group(P<0.05);The levels of E2 in the moderate-and high-fluoride groups were significantly lower than that in the control group(P<0.05);Significantly lower levels of BGP,BALP,IGF-1 and IGFBP5,higher levels of TRACP-5 b and IGFBP4 were observed in each exposure group compared with those in the control group(P<0.05).Conclusion Chronic coal-burning fluorosis-induced damage of E2 secretion in female rats may aggravate fluorosis,and the IGF family may be involved in this process.
