摘　　要：目的:建立天麻醒脑胶囊中天麻素、对羟基苯甲醇、毛蕊花糖苷、松果菊苷、梓醇、异毛蕊花糖苷、远志口山酮Ⅲ、3,6’-二芥子酰基蔗糖8种成分的含量测定方法,为天麻醒脑胶囊的内在质量控制提供实验参考。方法:采用HPLC波长切换法同时测定制剂中天麻素、对羟基苯甲醇、毛蕊花糖苷、松果菊苷、梓醇、异毛蕊花糖苷、远志口山酮Ⅲ、3,6’-二芥子酰基蔗糖8种化学成分的含量,色谱柱为Thermo BDS-C18柱(250 mm×4.6 mm,5μm);流动相为乙腈-0.1%磷酸水溶液,梯度洗脱;柱温为35℃;检测波长分别为λ1=220 nm、λ2=330 nm。结果:天麻素、对羟基苯甲醇、毛蕊花糖苷、松果菊苷、梓醇、异毛蕊花糖苷、远志口山酮Ⅲ、3,6’-二芥子酰基蔗糖分离度良好;分别在2.340~46.80μg/mL(r=0.999,9)、3.599~71.97μg/mL(r=0.999,8)、2.091~41.81μg/mL(r=0.999,8)、2.054~41.08μg/mL(r=0.999,7)、2.764~55.27μg/mL(r=0.999,9)、2.597~51.94μg/mL(r=0.999,9)、2.789~55.78μg/mL(r=0.999,8)、2.990~59.80μg/mL(r=0.999,9)的范围内线性关系良好;平均回收率分别为99.82%(RSD=1.10%,n=9)、100.79%(RSD=1.20%,n=9)、100.07%(RSD=0.43%,n=9)、101.22%(RSD=1.23%,n=9)、100.99%(RSD=1.31%,n=9)、98.98%(RSD=1.51%,n=9)、98.79%(RSD=1.02%,n=9)、97.83%(RSD=1.23%,n=9)。结论:该法同时定量分析了8种化学成分的含量,该法操作快速、准确、精密度高且重复性好,可用于天麻醒脑胶囊内在质量的有效评价。Objective:To establish a method for the determination of gastrodin,4-hydroxybenzyl alcohol,verbascoside,echinacosid,catalpol,isoacteoside,polygalaxanthoneⅢ,and 3,6’-disinapoyl sucrose in Tianma Xingnao capsule,and to provide an experimental reference for the internal quality control of Tianma Xingnao capsule.Methods:The contents of gastrodin,4-hydroxybenzyl alcohol,verbascoside,echinacosid,catalpol,isoacteoside,polygalaxanthoneⅢ,and 3,6’-disinapoyl sucrose inTianma Xingnao capsule were detected by HPLC with wavelength switching combined gradient elution method.The column was a Thermo BDS-C18 column(250 mm×4.6 mm,5μm);the mobile phase was acetonitrile-0.1%phosphoric acid aqueous solution,gradient elution;the column temperature was 35℃;the detection wavelength wereλ1=220 nm,λ2=330 nm.Results:The gastrodin,4-hydroxybenzyl alcohol,verbascoside,echinacosid,catalpol,isoacteoside,polygalaxanthoneⅢ,and3,6’-disinapoyl sucrose displayed good linear relationships with in the range of 2.340-46.80μg/mL(r=0.999,9),3.599-71.97μg/mL(r=0.999,8),2.091-41.81μg/mL(r=0.999,8),2.054-41.08μg/mL(r=0.999,7),2.764-55.27μg/mL(r=0.999,9),2.597-51.94μg/mL(r=0.999,9),2.789-55.78μg/mL(r=0.999,8),2.990-59.80μg/mL(r=0.9999),and the average recoveries were 99.82%(RSD=1.10%,n=9),100.79%(RSD=1.20%,n=9),100.07%(RSD=0.43%,n=9),101.22%(RSD=1.23%,n=9),100.99%(RSD=1.31%,n=9),98.98%(RSD=1.51%,n=9),98.79%(RSD=1.02%,n=9),97.83%(RSD=1.23%,n=9),respectively.Conclusion:The contents of 8 components were simultaneous detected.The method is simple,rapid,accurate and reproducible.It can be used as an effective method to evaluate the internal quality of Tianma Xingnao capsule.
