摘 要:目的:基于网络药理学探讨淫羊藿治疗多囊卵巢综合征的靶点和机制。方法:中药系统药理学分析平台(TCMSP)筛选淫羊藿活性成分及靶基因,GeneCards数据库获取多囊卵巢综合征相关基因,Venny将淫羊藿靶基因与PCOS相关基因映射,Cytoscape3.2.1软件构建淫羊藿-成分-靶点-疾病网络,STRING10.5软件构建PPI网络互作图和Origin9.1软件对PPI网络中的前30位蛋白绘制条形图,最后使用David v 6.8数据库将淫羊藿治疗多囊卵巢综合征的共同靶点做GO功能富集分析及KEGG通路富集分析。结果:共得到淫羊藿候选药效成分23个,涉及作用靶点106个,多囊卵巢综合征相关基因3739个,淫羊藿治疗多囊卵巢综合征的关键化合物21个,共同作用靶点67个。GO功能富集主要包含序列特异性DNA结合、转录因子结合、RNA聚合酶Ⅱ启动子转录的正调控等。KEGG富集通路主要有PI3K-AKt信号通路、TNF信号通路等72条。结论:筛选和分析淫羊藿治疗多囊卵巢综合征的作用机制,为后续进行深入研究提供了一定思路和参考。Objective:To explore the target and mechanism of Yinyanghuo(Epimedium)in the treatment of polycystic ovary syndrome(PCOS)based on network pharmacology.Methods:The active ingredients and target genes of Yinyanghuo(Epimedium)were screened through TCMSP,and the genes related to PCOS was obtained by GeneCards database.Venny mapped the target genes of Yinyanghuo(Epimedium)with PCOS.Yinyanghuo(Epimedium)-ingredients-target-disease network was built by Cytoscape 3.2.1 software.PPI network interaction diagram was built through STRING10.5 software and the first 30 proteins in PPI network were screened by Origin 9.1 software.Finally,the common target of Yinyanghuo(Epimedium)in the treatment of PCOS of GO function analysis and KEGG enrichment analysis were analyzed by David v 6.8 database.Results:A total of 23 candidate pharmacodynamic ingredients were obtained,involving 106 targets,3739 genes related to PCOS,21 key compounds in the treatment of PCOS and 67 common targets.GO function analysis mainly involves sequence-specific DNA binding,transcription factor binding and positive regulation of transcription from RNA polymeraseⅡpromoter.There are 72 KEGG enrichment analysis,mainly including P13 K-AKt signaling pathway and TNF signaling pathway.Conclusion:Screening and analyzing the mechanism of Yinyanghuo(Epimedium)in the treatment of PCOS provides some ideas and references for further study,which promotes the modernization of traditional Chinese medicine.
