摘 要:为检疫不同感染率貂场水貂阿留申病(AD)的感染状态,本研究利用碘凝集(IAT)和对流免疫电泳(CIEP)方法对水貂阿留申病毒(AMDV)不同感染率的两个貂场(A、B两个貂场)进行AD检疫,采用PCR和荧光定量PCR(qPCR)方法进一步分析AMDV的感染状态。结果显示,根据IAT和CIEP检测结果可将貂群分为4个群体:IAT-CIEP-、IAT+CIEP+、IAT-CIEP+和IAT+CIEP-,不同AMDV感染率貂场各群体占比有明显差异;A场母貂感染率显著高于公貂(p<0.01),而B场公貂与母貂感染率差异不显著。PCR结果表明,低感染率A貂场中IAT-CIEP-貂群中存在感染初期貂(IAT-CIEP-PCR+);高感染率B貂场中存在超过40%的抗性貂(IAT-CIEP+PCR-)和耐受貂(IAT-CIEP+PCR+)。qPCR结果显示,A场貂群的病毒载量集中分布在1.1×104拷贝/μL^7.5×105拷贝/μL,而B场貂群的病毒载量集中分布在1.8×103拷贝/μL^9.5×103拷贝/μL范围内。统计学分析显示,低感染率A貂场水貂体内AMDV载量显著高于高感染率的B貂场(p<0.01)。表明上述联合检疫方法可准确判断AD的感染状态,且不同感染率貂场AD的感染状态存在明显差异。本研究为貂场检疫淘汰阿留申病貂提供了参考依据。To analyze the infection of Aleutian disease(AD)in two mink farms,this study used the combination of iodine agglutination test(IAT)and counter immuno-electrophoresis(CIEP)to detect the infection of Aleutian mink disease virus(AMDV)in minks of two farms.Next,both PCR and real-time fluorescent quantitative PCR(qPCR)assays were used to further analyze the disease progression of minks infected with AMDV.The results showed that the mink herds could be divided into four groups:IAT-CIEP-,IAT+CIEP+,IAT-CIEP+,and IAT+CIEP-.The proportion of four groups in the two mink farms had a significant difference in AMDV infection rates.The PCR results indicated that there were some minks during early infection period(i.e.IAT-CIEP-PCR+)in the A farm with low AMDV infection rate;and more than 40%AMDV resistant minks(IAT-CIEP+PCR-)or AMDV tolerant minks(IAT-CIEP+PCR+)in the B farm with high AMDV infection rate.The qPCR result showed that the virus load in the A farm was 1.1×104 copies/μL-7.5×105 copies/μL,while that in the B farm was 1.8×103 copies/μL-9.5×103 copies/μL.Statistical analysis showed that virus load in minks from the A farm were much higher than minks from the B farm.In conclusion,the present study indicates that the farms with distinct AMDV infection rates have different distribution of mink infection status.The combined quarantine method can accurately determine the infection status of AD,which can provide a theoretical basis to better control AD in mink farm.
