摘 要:为探究炎症反应中东北白鹅Toll样受体15(TLR15)的免疫功能及其组织表达特征,本研究采用荧光定量PCR方法,对LPS诱导下的东北白鹅TLR15及相关免疫因子在不同组织的转录水平进行分析;同时利用免疫组织化学技术,对TLR15在不同组织中的表达水平进行检测。结果显示,在LPS诱导下东北白鹅TLR15及其相关免疫因子(IFN-α、IFN-γ、IL-1β和IL-6)的转录水平均高于对照组,其中TLR15、IFN-α和IFN-γ在法氏囊组织转录水平最高(p<0.01),IL-6在胸腺中转录水平最高,表明在炎症反应中TLR15及其免疫因子在不同组织的转录水平存在一定的差异。免疫组织化学检测结果显示,与对照组比较,各实验组TLR15在法氏囊、脾脏和肝脏中的表达差异极显著(p<0.01),在肺组织和肾组织中的表达差异显著(p<0.05),表明在LPS诱导下,鹅TLR15基因的转录和翻译水平一致。本研究为东北白鹅TLR15在炎症反应中的免疫功能分析提供了参考依据。Toll-like receptor 15(TLR15)is an important innate immune factor for the recognition of pathogen-associated molecular patterns(PAMPs).In this study,mRNA levels of TLR15 and related immune factors were detected by using real-time PCR in different tissues from Northeast white goose induced by bacterial lipopolysaccharide(LPS).The expression level of TLR15 in different tissues was analyzed by immunohistochemistry.The results showed that mRNA levels of TLR15 and immune factors(IFN-α、IFN-γ、IL-1βand IL-6)were higher in the experimental groups than those in the control group.For the experimental groups,the mRNA levels of TLR15,IFN-αand IFN-γgenes were the highest in the bursal tissue(p<0.01),and that of IL-6 was highest in spleen.The results of immunohistochemistry showed that the expression levels of TLR15 were significantly different in bursa,spleen and liver(p<0.01),and it was different(p<0.05)in lung and kidney.This is indicated that the transcription and translation of the goose TLR15 gene were at the same level under LPS induction.This study provides an essential foundation for the immune functional analysis of TLR15 in Northeast white goose in the inflammatory response.
