摘 要:为进一步探明本实验室前期从澳洲墨瑞鳕溃疡组织中首次分离的寄鱼不动杆菌(Acinetobacter piscicola)LW15的致病性和药敏特性,本实验再次明确了该菌株的系统进化地位,同时进行人工回归感染试验检测其致病性。结果显示菌株LW15的16SrDNA与Acinetobacter guillouiae CIP63.46T同源性最高,为97.7%,与本实验室前期分离的系同一菌株,为寄鱼不动杆菌;动物回归实验结果显示该菌为澳洲墨瑞鳕致病菌,半数致死浓度为4.26×106 cfu/mL。基于生物信息学方法对该分离菌进行毒力基因分析,结果显示其携带224个毒力基因,其中invasion、adherence和toxin等3类毒力基因中分别包含2、43和3个基因。对LW15病原菌进行药物敏感性检测,结果显示该菌株对氧氟沙星、环丙沙星、卡那霉素、庆大霉素、头孢曲松、丁胺卡那、哌拉西林等21种药物敏感,对羧苄西林和呋喃唑酮2种药物中度敏感,对四环素、克林霉素、头孢拉定、头孢氨苄、苯唑西林等7种药物不敏感。本研究为由寄鱼不动杆菌引起的澳洲墨瑞鳕溃疡疾病有效防治提供参考依据和实验数据。In order to further investigate the pathogenicity and antibiotic sensitivity of Acinetobacter piscicola LW15 previously isolated from diseased farmed Murray cod by our laboratory,in this study,the evolutionary position of strain LW15 was again verified,meanwhile,the tests on artificial regression infection were performed.The results indicated that strain LW15 shared the highest 16S rDNA homology to Acinetobacter guillouiae CIP 63.46T(97.7%),which was identical to the strain of Acinetobacter piscicola LW15.Strain LW15 was the pathogen for Murray cod and had the semi-lethal concentration of 4.26×106 cfu/mL.The virulence genes of stain LW15 was analyzed by the method of bioinformatics and its sensitivity to antibacterial drugs were performed.the results showed that the genome carried 224 virulence genes,of which invasion,adherence and toxin contained 2,43 and 3,respectively.In addition,strain LW15 isolate was sensitive to 21 antibiotics,such as ofloxacin,ciprofloxacin,kanamycin,gentamicin,ceftriaxone,butylamine and kanapacillin,etc.;and moderately sensitive to carbenicillin and furazolidone,but not sensitive to 7 antibiotics,such as tetracycline,clindamycin,cefradine,cephalexin or oxacillin.This study provides theoretical basis and experimental datails meaningful for the effective prevention and treatment of Murray cod disease caused by Acinetobacter spp.
