摘  要:目的:比较研究呋喃芬太尼与芬太尼的镇痛效应。方法:通过热板法对芬太尼和呋喃芬太尼的小鼠镇痛作用进行实验,采用皮下注射方式,观察不同剂量的芬太尼和呋喃芬太尼小鼠在热板上的舔后足时间,得出两种物质的镇痛效应曲线及半数有效剂量。结果:热板试验结果显示皮下注射5分钟后,芬太尼镇痛效应的ED50为0.08 mg/kg,呋喃芬太尼镇痛效应的ED50为0.13 mg/kg,为芬太尼效价的0.62倍。低剂量的芬太尼(0.02 mg/kg)与呋喃芬太尼(0.03 mg/kg)单次给药均可诱发小鼠短时间的痛觉过敏。结论:呋喃芬太尼与芬太尼一样,可剂量依赖性地对小鼠产生明显的镇痛作用,但镇痛作用强度和持续时间均弱于芬太尼。首次发现芬太尼与呋喃芬太尼在低剂量单次给药时能诱发小鼠短时间的痛觉过敏,具体作用机制还有待于进一步研究。Objective:The compare the analgesic effects of fentanyl and furanyfentanyl.Methods:In this paper,the analgesic effects of fentanyl and furanyfentanyl were tested by the hot plate antinociceptive method.Subcutaneous injections of fentanyl and furanyfentanyl at different doses were used to observe the time of licking rear feet of mice on the hot plate.Results:After 5 min of subcutaneous injections,the analgesic effect ED50 of fentanyl was 0.08 mg/kg and furanyfentanyl was 0.13 mg/kg,which was 0.62 times with fentanyl.In addition,single dose of 0.02 mg/kg fentanyl and 0.03 mg/kg furanyfentanyl could cause the phenomenon of acute hyperalgesia.Conclusion:Similar to fentanyl,furanyfentanyl has a dose-dependent analgesic effect on mice,but the intensity and duration of analgesic effects are weaker than that of fentanyl.For the first time,it was found that low dose of fentanyl and furanyfentanyl could induce a short period of hyperalgesia in mice when given in a single dose.The specific mechanism of action remains to be further studied.
