摘 要:目的考察SBA-15、MCM-41这两种型号的介孔二氧化硅载体载艾塞那肽的处方工艺以及比较两者载药性能差异和体外释药特性。方法采用单因素考察结合正交试验,以两种载体载艾塞那肽的载药量为评价指标,进行处方工艺的优化,采用透析袋法测定两种载药纳米粒的体外释药特性。结果 SBA-15载艾塞那肽的优化处方为温度50℃、TEOS用量10mL、P-123的用量3mL,测得平均载药量为14.93%;MCM-41载艾塞那肽的优化处方为温度80℃、CTAB浓度0.3%、TEOS的用量1.7mL,测得平均载药量为14.63%,两种载药纳米粒均呈现明显的缓释特性。结论相比于MCM-41载体,SBA-15载体具有相对大的载药量,且稳定性良好。OBJECTIVE To investigate the preparation process of two kinds of mesoporous silica nanoparticles MCM-41 and SBA-15 to load exenatide(EXT).The drug loading abilities andin vitro drug release characteristics of the two kinds of carries were compared.METHODS The study was carried out with single factor investigation and orthogonal experiment and the drug loading efficiencywas used as the index of inclusion process.The in vitro drug release characteristics were investigated by dialysis bag method.RESULTS The optimal conditions of EXT loaded SBA-15 were:temperature:50℃,TEOS:9 mL,P-123:3 mL,the average drug loading efficiency was 14.93%;the optimal conditions of EXT loaded MCM-41 were:temperature:80℃,CTAB:0.3%,TEOS:1.7 mL,the average drug loading efficiency was 14.63%.CONCLUSION SBA-15 is more feasible to load EXT than MCM-41.
