摘 要:目的建立HPLC法测定人血中伏立康唑浓度的方法,并应用于患者的个体化治疗。方法以Agilent technologies ZORBAX SB-C18(150×4.6mm,5μm)色谱柱分离,柱温30℃,流动相为甲醇∶水=53∶47,流速1.0mL·min-1,检测波长为256nm,进样量为30μL,卡马西平为内标。结果伏立康唑浓度在0.31~19.84μg·mL-1范围内,呈良好的线性关系,最低检测限0.31μg·mL-1(S/N≥10)。结论本方法快速、简便、稳定,适合用于伏立康唑血药浓度的测定。伏立康唑血药浓度个体差异大,开展治疗药物监测有利于保证患者安全、有效用药。OBJECTIVE Establish a method for the determination of voriconazole in human blood by HPLC and apply it to individualized treatment of patients.METHODS Sample was separated by Agilent technologies ZORBAX SB-C18(150×4.6 mm,5μm)column,column temperature was 30℃.The mobile phase was as follows:methanol∶water was 53∶47,the flow rate was 1.0 mL·min-1,the detection wavelength was 256 nm,injection volume was 30μL.Carbamazepine was an internal standard.RESULTS Range of linearity of voriconazole was 0.31-19.84μg·mL-1.Minimum detection limit was 0.31μg·mL-1(S/N≥10).CONCLUSION The method is rapid,simple and stable,and is suitable for the determination of voriconazole blood concentration.The blood concentration of voriconazole varies greatly,and the monitoring of therapeutic drugs is beneficial to ensure the safe and effective use of drugs.
