摘  要:目的探讨硼替佐米对重症急性胰腺炎(SAP)犬血浆炎性细胞因子及肠道屏障功能的影响。方法用自身胆汁逆行主胰管注射法建立SAP模型,并随机分为模型组、实验组及对照组,每组7只;另取7只健康成年杂种犬作为空白组,仅暴露主胰管后关闭伤口。模型组及空白组均给予0.9%NaCl 1 mL皮下注射,q12 h;实验组给予注射用硼替佐米0.15 mg·kg^-1皮下注射,qd;对照组给予醋酸奥曲肽注射液0.1 mg皮下注射,q12 h。4组犬均给药5 d。用全自动酶联免疫工作站检测血浆炎症细胞因子水平,用鲎试剂基质显色定量法测定血浆内毒素(LPS)水平。结果实验组、对照组、模型组的肿瘤坏死因子-α分别为(115.74±15.83),(154.82±21.57)和(429.63±61.29)pg·mL^-1,白细胞介素-6(IL-6)分别为(141.72±18.94),(177.56±22.19)和(417.34±50.06)pg·mL^-1,IL-1β分别为(118.65±15.10),(137.24±16.38)和(162.87±21.19)pg·mL^-1,LPS分别为(1.79±0.25),(2.01±0.27)和(2.58±0.36)EU·mL^-1,实验组的上述指标与模型组和对照组比较,差异均有统计学意义(均P<0.05)。结论硼替佐米能有效地降低SAP犬的血浆细胞因子水平,减轻胰腺组织病理损伤程度。Objective To analyze the effects of bortezomib on plasma inflammatory cytokines and intestinal barrier function in dog model with severe acute pancreatitis(SAP).Methods SAP model was established by autologous bile retrograde main pancreatic duct injection,and the model dogs were randomly divided into model group,experimental group and control group with 7 dogs per group.Another 7 healthy adult mongrel dogs were taken as the blank group,and only the main pancreatic duct was exposed and then the wound was closed.Model group and blank group were given 0.9%NaCl 1 mL by subcutaneous injection,q12 h.Experimental group was given bortezomib injection 0.15 mg·kg^-1 mL by subcutaneous injection,qd.Control group was given octreotide acetate injection 0.1 mg by subcutaneous injection,q12 h.Four groups were treated for 5 days.The plasma inflammatory cytokines levels were detected by the automatic enzyme-linked immunosorbent workstation,the plasma endotoxin(LPS)were detected by the limulus amebocyte lysate substrate colorimetry.Results The levels of plasma tumor necrosis factor-αin experimental,control and model groups were(115.74±15.83),(154.82±21.57)and(429.63±61.29)pg·mL^-1,interleukin-6(IL-6)were(141.72±18.94),(177.56±22.19)and(417.34±50.06)pg·mL^-1,IL-1βwere(118.65±15.10),(137.24±16.38)and(162.87±21.19)pg·mL^-1,LPS were(1.79±0.25),(2.01±0.27)and(2.58±0.36)EU·mL^-1.The above indexes of the experimental group were significantly different from those in model and control groups(all P<0.05).Conclusion Bortezomib can effectively reduce the plasma cytokine levels in dogs with SAP,and reduce the pathological injury degree of pancreatic tissue.
