摘  要:目的研究重组人促红细胞生成素(rhE-PO)对肾缺血再灌注损伤(RIRI)大鼠肾组织Nod样受体蛋白3(NLRP3)表达的影响。方法SD大鼠通过阻断左侧肾血液供应构建RIRI模型,假手术组不阻断左侧肾血液供应,将建模成功的RIRI大鼠随机分为模型组和实验组各15只。实验组于RIRI建模后腹腔注射5000 U·kg^-1 rhE-PO,假手术组和模型组给予等量生理盐水。以生化分析仪检测各组RIRI大鼠血清肾功能指标,以酶联免疫吸附法检测各组大鼠肾组织中炎性因子含量,以免疫组化检测大鼠肾组织Nod样受体蛋白3(NLRP3)蛋白表达。结果假手术组、模型组和实验组大鼠血清肌酸酐分别为(32.93±17.56),(121.21±22.34),(89.49±18.45)mmol·L^-1,尿素氮分别为(8.76±1.25),(31.42±2.37),(23.55±1.62)mmol·L^-1,肾组织中肿瘤坏死因子-α分别为(0.54±0.11),(6.42±0.32),(3.65±0.18)pg·mg^-1,白细胞介素-1β分别为(1.36±0.15),(5.69±2.04),(2.31±0.11)pg·mg^-1,肾组织中NLRP3蛋白表达分别为(5.36±2.18)%,(56.28±3.45)%,(16.42±5.04)%,差异均有统计学意义(均P<0.05)。结论rhE-PO可通过调控肾组织NLRP3蛋白表达改善RIRI大鼠肾功能,减轻肾组织炎性损伤。Objective To investigate the effect of recombinant human erythropoietin(rhE-PO)on the expression of Nod-like receptor protein 3(NLRP3)in renal tissue of rats with renal ischemia-reperfusion injury(RIRI).Methods The RIRI model of SD rats was established by blocking the blood supply of the left kidney.The sham-operation group did not block the blood supply of the left kidney.The RIRI rats were randomly divided into model group and test group,with 15 rats in each group.Test group was intraperitoneal injected with 5000 U·kg^-1 rhE-PO after RIRI modeling,sham-operation group and model group were given the same amount of normal saline.Biochemical analyzer was used to detect the serum renal function indexes of RIRI rats.Enzyme-linked immunosorbent assay was used to detect the content of inflammatory factors in the kidney tissues of rats in each group.The expression of NLRP3 in the kidney tissues of rats was detected by immunohistochemistry.Results The creatinine in sham-operation group,model group and test group were(32.93±17.56),(121.21±22.34),(89.49±18.45)mmol·L^-1,serum urea nitrogen were(8.76±1.25),(31.42±2.37),(23.55±1.62)mmol·L^-1,tumor necrosis factor-αin renal tissue were(0.54±0.11),(6.42±0.32),(3.65±0.18)pg·mg-1,interleukin-1βin renal tissue were(1.36±0.15),(5.69±2.04),(2.31±0.11)pg·mg-1,the expression of NLRP3 protein in kidney tissue were(5.36±2.18)%,(56.28±3.45)%,(16.42±5.04)%,all with significant difference(all P<0.05).Conclusion RhE-PO can improve renal function and alleviate inflammatory injury in RIRI rats by regulating the expression of NLRP3 protein in renal tissue.
