摘 要:目的研究柴胡皂苷-b2(SS-b2)对二乙基亚硝胺(DEN)诱导的原发性肝癌小鼠血管生成的影响。方法雄性BALB/c小鼠随机分为空白组、模型组、对照组(阿霉素1 mg·kg^-1)和低、中、高剂量实验组(SS-b21.5,3,6 mg·kg^-1),腹腔注射给药。实验结束时摘取肝、脾、胸腺称重,计算脏器指数;以Western Blot法检测肝血管内皮生长因子(VEGF)、基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)蛋白的表达变化。结果与空白组相比,模型组的肝指数、胸腺指数、脾指数和白细胞计数均明显升高(均P<0.01);与模型组比较,高剂量实验组肝指数、胸腺指数、脾指数和白细胞计数均显著降低(均P<0.01)。Western Blot结果显示,模型组肝VEGF,MMP-2和MMP-9的表达分别为1.66±0.10,1.12±0.08和2.21±0.36,高剂量实验组分别为0.36±0.04,0.25±0.04和0.77±0.21,差异均有统计学意义(均P<0.01)。结论柴胡皂苷-b2对原发性肝癌的发生发展具有一定的抑制作用,其机制可能与其下调血管生成相关蛋白VEGF,MMP-2,MMP-9的表达有关。Objective To study the effect of Saikosaponin-b2(SS-b2)on angiogenesis in mice with primary liver cancer induced by diethylnitrosamine(DEN).Methods Male BALB/c mice were randomly divided into blank group,model group,control group(doxorubicin hydrochloride 1 mg·kg^-1)and low,medium,high dose experimental groups(SS-b21.5,3,6 mg·kg^-1),intraperitoneal injection.At the end of the experiment,the liver,spleen and thymus were removed and weighed,the organs index were calculated.The protein expressions of vascular endothelial growth factor(VEGF),matrix matalloproteinases-2(MMP-2)and matrix matalloproteinases-9(MMP-9)in liver were detected by Western Blot.Results Compared with blank group,the liver index,thymus index,spleen index and white blood cell counts were significantly increased in model group(all P<0.01).Compared with model group,the liver index,thymus index,spleen index,and white blood cell count in high dose experimental group were significantly reduced(all P<0.01).Western Blot showed that the expressions of VEGF,MMP-2 and MMP-9 in liver tissue of model group were 1.66±0.10,1.12±0.08,2.21±0.36,high dose experimental group were 0.36±0.04,0.25±0.04,0.77±0.21,all with significant difference(all P<0.01).Conclusion SS-b2 inhibited the development of primary liver cancer,and its mechanism may be related to down-regulating the expressions of angiogenesis associated protein VEGF,MMP-2 and MMP-9.
