摘  要:目的探讨17β雌二醇对重复吸入七氟烷导致的新生大鼠认知障碍的影响。方法将新生SD大鼠随机分为对照组、模型组和实验组,模型组和实验组分别于7,14,21日龄吸入2.6%七氟烷4 h;对照组仅吸入氧气4 h;每次麻醉吸入前20 min,实验组腹腔注射500μg·kg^-117β雌二醇,模型组和对照组腹腔注射等量生理盐水。以电子天平称重各组大鼠7,14,21日龄时脑质量的变化,以Morris水迷宫实验检测大鼠30日龄(幼年期)及90日龄(成年期)学习记忆功能,以免疫组化法检测大鼠海马组织缝隙连接蛋白(Cx30)表达情况,以蛋白质印迹法检测大鼠海马组织中脑源性神经营养因子(BDNF)、白细胞介素-6(IL-6)蛋白的表达。结果对照组、模型组及实验组幼年期大鼠定位航行实验第5天的逃避潜伏期为(15.84±4.58),(27.67±5.34),(17.79±3.15)s,跨台次数分别为(14.26±3.25),(7.25±1.26),(12.26±3.28)次,成年期大鼠定位航行实验第5天的逃避潜伏期为(12.63±2.48),(25.36±4.15),(14.25±2.09)s,跨台次数分别为(15.48±2.79),(9.53±2.38),(13.53±3.39)次。与对照组比较,模型组及实验组幼年期及成年期大鼠的逃避潜伏期均延长,跨台次数减少,实验组大鼠的逃避潜伏期均较模型组缩短,跨台次数增加(P<0.05)。对照组、模型组和实验组大鼠海马组织Cx30蛋白、BDNF蛋白相对表达量和IL-6蛋白相对表达量,组间比较差异均有统计学意义(均P<0.05)。结论17β雌二醇可改善重复吸入七氟烷导致的新生大鼠近期及远期认知障碍,调控海马组织中BDNF、IL-6蛋白表达进而降低对脑组织的损伤。Objective To investigate the effect of 17βestradiol on cognitive impairment in neonatal rats induced by repeated sevoflurane inhalation.Methods Newborn SD rats were divided randomly into control group,model group and test group.Model group and test group inhaled 2.6%sevoflurane for 4 h at the age of 7,14 and 21 d respectively;control group only inhaled oxygen for 4 h;20 mins before each anesthesia inhalation,test group was intraperitoneal injection with 500μg·kg^-117βestradiol,model group and control group were intraperitoneal injection with the same amount of normal saline.The changes of brain mass at the age of 7,14 and 21 days in each group were weighed by electronic balance.Morris water maze test was used to detect the learning and memory function of rats at the age of 30 days(in infancy)and 90 days(in adulthood).Immunohistochemical method was used to detect the expression of connexons 30(Cx30)in the hippocampus of rats.Western blotting was used to detect the expression of brain-derived neurotrophic factor(BDNF)and interleukin-6(IL-6)protein in the hippocampus of rats.Results The escape latency in control group,model group and test group of young rats were(15.84±4.58),(27.67±5.34),(17.79±3.15)s on the 5th day of the positioning navigation experiment,the number of platform crossing were 14.26±3.25,7.25±1.26,12.26±3.28,and the escape latency of adult rats were(12.63±2.48),(25.36±4.15),(14.25±209)s,the number of platform crossing were 15.48±2.79,9.53±2.38,13.53±3.39.Compared with control group,the escape latency in model group and test group of young rats and adult rats prolonged of the positioning navigation experiment,and the number of cross platform decreased.The escape latency of rats in test group was shorter than that in model group,and the number of crossing stations increased(P<0.05).The percentage of Cx30 protein positive cells in hippocampus,the relative expression of BDNF protein,the relative expression of IL-6 protein in control group,model group and test group were all had statistica
