摘 要:目的研究18β-甘草次酸(18β-GA)对变应性鼻炎(AR)大鼠鼻黏膜重塑及鼻黏膜水通道蛋白1(AQP1)、胸腺基质淋巴细胞生成素(TSLP)表达的影响。方法将SD大鼠120只,随机分为空白组(n=30)、模型组(n=30)、实验组(n=30)及对照组(n=30)。模型组、实验组及对照组大鼠均用卵清蛋白(OVA)诱导致敏的方法建立AR大鼠模型。造模成功后,实验组大鼠灌胃18β-GA(25 mg·kg^-1·d^-1),对照组大鼠灌胃氯雷他定(1 mg·kg^-1·d^-1),空白组与模型组灌胃等量生理盐水,连续干预8周。用叠加量化记分法记录各组大鼠搔鼻、喷嚏、鼻溢液症状行为;用酶联免疫吸附(ELISA)法检测血清OVA特异性IgE(OVA-sIgE)、转化生长因子-β1(TGF-β1)和白细胞介素-17(IL-17)水平;用蛋白质印迹(Wb)法检测鼻黏膜组织AQP1、TSLP蛋白表达。结果空白组、模型组、实验组及对照组大鼠血清OVA-sIgE水平分别为(7.32±2.46),(21.43±5.62),(16.73±3.67),(12.85±4.06)ng·mL-1;TGF-β1水平分别为(8.78±3.43),(23.84±4.38),(18.11±4.93),(14.37±5.02)ng·mL-1;IL-17水平分别为(552.43±41.32),(782.61±70.21),(664.72±53.28),(595.87±46.63)ng·mL-1。与空白组相比,模型组以上各指标均显著升高,且实验组低于模型组,对照组低于实验组(均P<0.05)。结论18β-GA可抑制AR大鼠血清OVA-sIgE水平,同时抑制炎性介质的释放及鼻黏膜组织AQP1、TSLP蛋白表达,减轻鼻黏膜组织的炎性损伤,缓解过敏症状。Objective To investigate the effects of 18β-glycyrrhetinic acid(18β-GA)on nasal mucosal remodeling and expression of aquaporin-1(AQP1)and thymic stromal lymphopoietin(TSLP)in nasal mucosa of rats with allergic rhinitis(AR).Methods A total of 120 SD rats were randomly divided into blank group(n=30),model group(n=30),test group(n=30)and control group(n=30).Rats in model group,test group and control group were induced to sensitization by ovalbumin(OVA)to establish AR rat models.After successful modeling,test group was gavage with 18β-GA(25 mg·kg^-1·d^-1),control group was gavage with loratadine(1 mg·kg^-1·d^-1),blank group and model group were given the same amount of normal saline for 8 weeks.The symptoms of nasal,sneezing and nasal discharge in each group were recorded by superposition quantitative scoring method;serum OVA-specific IgE(OVA-sIgE),transforming growth factor-β1(TGF-β1)and interleukin-17(IL-17)levels were measured by enzyme-linked immunosorbent assay(ELISA);Western blotting(Wb)method was used to detect the expression of AQP1 and TSLP proteins in nasal mucosa.Results The serum OVA-sIgE levels of blank group,model group,test group and control group were(7.32±2.46),(21.43±5.62),(16.73±3.67),(12.85±4.06)ng·mL^-1,the levels of TGF-β1 were(8.78±3.43),(23.84±4.38),(18.11±4.93),(14.37±5.02)ng·mL^-1,the IL-17 levels were(552.43±41.32),(782.61±70.21),(664.72±53.28),(595.87±46.63)ng·mL^-1.Compared with blank group,the above indexes of model group were significantly increased,and test group were lower than model group,control group were lower than test group(all P<0.05).Conclusion 18β-GA can inhibit the serum OVA-sIgE level in AR rats,inhibit the release of inflammatory mediators and the expression of AQP1 and TSLP proteins in nasal mucosa,alleviate the inflammatory injury of nasal mucosa and relieve allergic symptoms.
