摘 要:目的观察鬼针草总黄酮(TFB)单次和重复给药39周口服对Beagle犬产生的毒性。方法根据最大合理给药容量以4只Beagle犬进行单次给药毒性实验;以低、中、高3个剂量实验组(0.1,0.3,1.0 g·kg^-1·d^-1 TFB)和对照组(空心胶囊)对40只Beagle犬进行39周经口重复给药的毒性实验,检测血液学、血液生化和病理组织学各项指标,评价TFB的安全性。结果TFB经口给予大于1.0 g·kg^-1·d-1时,Beagle犬出现明显胃肠道反应。单次给药毒性实验中,动物肝、肾均有不同程度损伤;重复给药毒性实验中,部分动物有肝、肾损伤,但无剂量效应关系。结论TFB具有较好的安全性,但临床使用应定期监测肝、肾功能指标。Objective To investigate the single dose and the repeated dose toxicity of total flavonoids from Bidens pilosa(TFB)in Beagle dogs.Methods The single dose toxicity test was conducted in 4 Beagle dogs with the reasonable maximum dose capacity.The repeated dose toxicity test was carried out on 40 Beagle dogs with low,middle,high dose experimental groups(TFB 0.1,0.3,1.0 g·kg^-1)and the control group(hollow capsule),for 39 weeks.The hematology,blood biochemistry and histopathology indexes were measured for toxicity assessment.Results Beagle dogs showed significant gastrointestinal adverse reactions when TFB was administered orally at least 1.0 g·kg^-1.Histological examination indicated that liver and kidney were damaged in the single dose toxicity test,some were injured in the repeated dose toxicity test.But there was no dose-response relationship.Conclusion TFB has better safety,but the liver and kidney function indexes should be monitored regularly during clinical use.
