摘 要:为了解H5N3亚型禽流感病毒(AIV)的生物学特性,本研究对从我国广西鸭体内分离到的一株低致病力H5N3亚型AIV DK/GX/S11453/2019(H5N3)进行了全基因组序列分析、抗原性分析、对小鼠的感染性以及受体结合特性分析。全基因组序列分析显示,该病毒的基因具有明显的野鸟源性,该病毒除HA与NA来源相同外,其余片段均来源不同,具有明显的遗传多样性。HA蛋白的裂解位点仅含有一个碱性氨基酸,符合低致病力AIV的分子特征。抗原性分析结果显示该病毒分离株与近几年国内使用的各疫苗株的抗原差异性较大。小鼠感染性试验结果显示,该病毒对小鼠呈现低致病力,不引起小鼠明显的体质量下降,但可以在小鼠的肺脏和鼻甲内有效的复制。受体结合特性分析结果显示,该病毒具有同时结合α-2,3和α-2,6受体的能力,存在一定感染哺乳动物的风险。本研究通过对该H5N3亚型AIV部分生物学特性的分析,为H5N3亚型禽流感的防控提供理论依据。To understand the biological characteristics of H5N3 avian influenza viruses(AIV),we analyzed the complete genome sequence,antigenicity,replication in mice and receptor binding properties of a H5N3 low pathogenic Avian influenza virus DK/GX/S11453/2019(H5N3)isolated from duck during routine surveillance in Guangxi province,China.Genomic sequence analysis revealed that the virus genes belonged to wild bird lineage.The HA and NA genes were most closely related to a same virus,whereas the others were phylogenetically diverse.The H5 virus possessed a single basic amino acid in the HA cleavage site,conforming to the signature of low-pathogenicity AIVs.Antigenicity analysis revealed that the antigens of this virus were quite different from those of other vaccine strains used in China in recent years.The results of the replication in mice showed that the virus could be detected in the lungs and nasal turbinates but did not cause significant body weight loss,indicating the low pathogenicity to mice.Receptor binding analysis demonstrated that the H5N3 subtype AIV possessed the ability to simultaneously bindα-2,3-linked glycans andα-2,6-linked glycans,thereby posing a potential threat to the health of mammals.Our study provides theoretical basis for prevention and control of H5N3 subtype avian influenza by analyzing part of biological characteristics of the H5N3 AIV.
